Project description:Purpose: The goal of this study is to reveal the transcriptional dynamics of hamster liver upon DC371739 treatment Methods: Total RNAs were extracted from the livers of HFD-hamsters control and DC371739-treated (30 mg/kg for 21 days) hamsters. Seven biological replicates were performed for each studied condition. The libraries were sequenced using NovaSeq 6000 (pair-end 150 bp). All reads were mapped to the reference genome of Mesocricetus auratus using HISAT2 v.2.2.1 as instructed. RNA counts were generated from feature Counts. Results: Ccompared with the vehicle group, the transcription levels of Pcsk9 and Angptl3 of the DC371739 treated group have been decreased by 41.3% (P = 0.016) and 15.7% (P = 0.031), respectively, while the transcription levels of Hnf1α and Ldlr have not shown the statistically significant difference after DC371739 treatment. Conclusions: We conclude that DC371739 negatively modulates the transcription of two crucial therapeutic targets PCSK9 and ANGPTL3 for hypercholesterolemia treatment.
