Inhibitors molecules isolated from midgut and midgut diverticula of the spider Nephilingis cruentata

Project description:Spiders are insect predators and their venoms are important to prey capture. Spider venomshave several potential applications as pharmaceutical compounds and insecticides. However,transcriptomic and proteomic analyses of the digestive system (DS) of spiders show that DS is also a rich source of new inhibitor molecules. Biochemical, transcriptomic and proteomic data of crude DS extracts show the presence of molecules with inhibitor potential in the spider Nephilingis cruentata. Therefore, the aims of this work were to isolate and characterize molecules with serine peptidase inhibitory activity. The DS of fasting adult females was homogenized under acidic conditions and subjected to heat treatment. After that, samples were submitted to ion exchange batch and high-performance reverse-phase chromatography. The fractions with inhibitory activity were confirmed by mass spectrometry, identifying 6 molecules with inhibitor potential. The inhibitor Nc6834 was kinetically characterized, showing a KD value of 30.25 nM ± 8.13. Analysis of the tertiary structure by molecular modeling using Alpha-Fold2 indicates that the inhibitor Nc6834 structurally belongs to theMIT1-like atracotoxin family. This is the first time that a serine peptidase inhibitory function is attributed to this structural family and the reactive site inhibitor residue is identified. Sequence analysis indicates that these molecules may be present in the DS of other spiders.

Spatial Variation of <i>Acanthophlebia cruentata</i> (Ephemeroptera), a Mayfly Endemic to Te Ika-a-Maui-North Island of Aotearoa, New Zealand.

Project description:The mayfly Acanthophlebia cruentata of Aotearoa, New Zealand, is widespread in Te Ika-a-Māui North Island streams, but has never been collected from South Island despite land connection during the last glacial maximum. Population structure of this mayfly might reflect re-colonisation after volcanic eruptions in North Island c1800 years ago, climate cycling or conceal older, cryptic diversity. We collected population samples from 33 locations to estimate levels of population genetic diversity and to document phenotypic variation. Relatively low intraspecific haplotype divergence was recorded among mitochondrial cytb sequences from 492 individuals, but these resolved three geographic-haplotype regions (north, west, east). We detected a signature of isolation by distance at low latitudes (north) but evidence of recent population growth in the west and east. We did not detect an effect of volcanic eruptions but infer range expansion into higher latitudes from a common ancestor during the last glacial period. As judged from wing length, both sexes of adult mayflies were larger at higher elevation and we found that haplotype region was also a significant predictor of Acanthophlebia cruentata size. This suggests that our mitochondrial marker is concordant with nuclear genetic differences that might be explained by founder effect during range expansion.
