The distant Microbial communities in Tibetan alpine meadow based on Geochip 4.0

Project description:Due to its high altitude and extreme climate conditions, the Tibetan plateau is a region vulnerable to the impact of climate changes and anthropogenic perturbation, thus understanding how its microbial communities function may be of high importance. Here, we report a study to profile soil microbial structural genes, which infers functional roles of microbial communities, aiming to explore potential microbial responses to climate changes and anthropogenic perturbation. Using a microarray-based metagenomics tool named GeoChip 4.0, we showed that microbial communities in treatment site were distinct, compared with those in control site, e.g. shrubland vs grassland, grazing site vs ungrazing site, or warmer site vs colder site. Substantial variations were apparent in stress, N and C cycling genes, but they were in line with the functional roles of these genes.

RNA-Seq data of Thalassiosira hyalina and Nitzschia frigida over a time course of high-light exposure under low and high pCO2

Project description:The diatoms Thalassiosira hyalina and Nitzschia frigida are important members of Arctic pelagic (open-water) and sympagic (ice-associated) microalgal communities. We investigated here the molecular mechanisms these algae apply to cope with abrupt high light exposure (Shift from 20 to 380 µmol photons m-2 s-1, resembling upwelling or ice break-up). Experiments were done under contemporary as well as future pCO2 (400 vs. 1000 µatm) to investigate whether [CO2] or pH modulate the reactions to high light intensities. After proper acclimation to the low-light regime, cells were sampled (= 0 hours), then exposed to the high-light treatment expression patterns were followed over 120h. Transcriptomic data were discussed also in the light of an accompanying physiological dataset from the same experiment (Ane Kvernvik et al. in prep).

Transcriptome of alfalfa plants under animal grazing

Project description:By comparing transcriptomes of tolerant and intolerant plants of sickle alfalfa subject to intensive animal grazing, we identified pathways involved in nutrient-responsive signaling, light and wound response, cell wall formation, and energy metabolism. In these pathways, grazing suppressed 39 genes, but less severe in the tolerant plant, and activated 5 genes all carrying polymorphisms in their homologous transcripts between the tolerant and intolerant plants. These genes and pathways - responsive to grazing and differentially expressed between the tolerant and intolerant plants â?? underline a defense mechanism in alfalfa against grazing stresses. We examined transcriptomes of 3 alfalfa plants: grazing tolerant and grazed, grazing tolerant and not grazed, grazing intolerant and grazed, all at single replicate each.

Project description:In order to investigate the diurnal oscillations of ruminal protozoa, and their responses to the changes in different feeding patterns, we conducted an animal experiment by feeding the sheep ad libitum with a hay-based diet (50% of alfalfa hay and 46% of oats hay) and a grain-based diet (45% of corn meal and 11% of soybean meal) for 30 days, and ruminal fluid samples were collected at six different timepoints from T2 to T22 in one day, and the composition and diversity of the protozoal communities in rumen microbiomes of the sheep in the Grain-diet and Hay-diet groups at different timepoints were analyzed through 18S rRNA sequencing.

Morphine treatment leads to temporal modulation of the gut microbiome

Project description:Opioid analgesics are frequently prescribed in the United States and worldwide. However, serious side effects such as addiction, immunosuppression and gastrointestinal symptoms limit long term use. In the current study using a chronic morphine-murine model a longitudinal approach was undertaken to investigate the role of morphine modulation of gut microbiome as a mechanism contributing to the negative consequences associated with opioids use. The results revealed a significant shift in the gut microbiome and metabolome within 24 hours following morphine treatment when compared to placebo. Morphine induced gut microbial dysbiosis exhibited distinct characteristic signatures profiles including significant increase in communities associated with pathogenic function, decrease in communities associated with stress tolerance. Collectively, these results reveal opioids-induced distinct alteration of gut microbiome, may contribute to opioids-induced pathogenesis. Therapeutics directed at these targets may prolong the efficacy long term opioid use with fewer side effects.
