Project description:Enlargement and doming of the shoot apical meristem (SAM) is a hallmark of the transition from vegetative growth to flowering. While this change is widespread, its role in the flowering process is unknown. The late termination (ltm) tomato mutant shows severely delayed flowering and precocious doming of the vegetative SAM. LTM encodes a kelch domain-containing protein, with no link to known meristem maintenance or flowering time pathways. LTM interacts with the TOPLESS (TPL) corepressor and with several transcription factors, providing specificity for its repressive functions. A sub-group of flowering-associated genes are precociously upregulated in vegetative stages of ltm SAMs, among them, the antiflorigen gene SELF PRUNING (SP). A mutation in SP restored the structure of vegetative SAMs in ltm sp double mutants and late flowering was partially suppressed, suggesting LTM functions to suppress SP in the vegetative SAM. In agreement, SP-overexpressing wild type plants exhibited precocious doming of vegetative SAMs combined with late flowering, as found in ltm plants. Strong flowering signals can result in termination of the SAM, usually by its differentiation into a flower. We propose that activation of a floral antagonist that promotes SAM growth in concert with floral transition protects it from such terminating effects.

Analysis of the mp clv3 transcriptome profile in Arabidopsis

Project description:Due to the major limitation of small set of stem cells located in the Shoot Apical Meristem (SAM), it is difficult to perform the whole genome analysis specific for those stem cells. Here, we generated the mp clv3 double mutant with a larger Shoot Apical Meristem,in which shoot stem cells were drastically enriched while the differentiation of stem cells was blocked. Then we performed the mRNA profiling on wild type and mp clv3 double mutant. And this could offer an ideal genetic material to highly increase the resolution of plant shoot stem cell profiling, also to further explore the mechanism of plant stem-cell regulation.

Analysis of the effect of auxin biosynthesis inhibitor L-AOPP on the gene expression profile (shoot apical meristem)

Project description:Recently, 2-aminoxy-3-phenylpropionic acid (L-AOPP) had been demonstrated to possess an inhibitory activity against IAA biosynthesis but the molecular basis of the action was unclear. To investigate the function of L-AOPP, we conducted microarray analysis using the shoot apical meristem (SAM) part of A. thaliana in addition to whole plants after the treatment of L-AOPP. We performed microarray analysis using the shoot apical meristem (SAM) part of A. thaliana (Columbia-0) to investigate function of 2-aminoxy-3-phenylpropionic acid (L-AOPP) in relation to inhibition of auxin biosynthesis. Total RNA was extracted from SAM part of 7-day-old seedlings grown on 1/2 MS medium containing L-AOPP (50 µM),L-AOPP (50 µM) + Indole-3-acetic acid (IAA 10 nM) or Dimethyl sulfoxide (DMSO 0.1%).

Proteomic analysis of the apoplast reveals a differential cold acclimation response among winter wheat crown tissues

Project description:The crown is the critical region for survival of winter wheat exposed to low temperature stresses. When wheat is exposed to non-freezing low temperatures, they can increase their freezing tolerance (cold acclimation, ACC). Changes within the apoplast are thought to be crucial for acquisition of freezing tolerance. However, how individual tissues within the ccrown, namely the shoot apical meristem (SAM, responsible for new shoot growth) and vascular transition zone (VTZ, located at the base of the crown)enhance tolerance to freezing has not yet been characterized. In the present study, we conducted shotgun proteomic analysis of the apoplast fluid to investigate ACC-induced proteins in the SAM and VTZ.
