Project description:We observed the effects of TDAG8-overexpression in Lewis lung carcinoma (LLC) cells on the gene expression pattern. TDAG8-overexpressing or vector-transfected control LLC cells (5 × e5 in 200 µL of HBSS) were injected into the lateral tail veins of 6–8-week-old C57BL/6 mice. Lungs were removed on day 19 post-injection, and total RNA was extracted.

Project description:Assay of gene expression pattern differences between liver cancer tissue and normal liver tissue from the same mouse by microarray in 4 separate mice injected with recombinant adeno-associated viral (AAV) vector 4 Mice (M24, M48, M50, M60) were necropsied at 18 months post-injection of AAV-vector, and livers removed. Tumor tissue and adjacent normal tissue were identified and dissected for RNA extraction.

A truncated reverse transcriptase enhances prime editing by split AAV vectors

Project description:To investigate if the truncated PE can be dilivered by dual AAV8 vectors for in vivo prime editing. We injected the dual AAV8 into 10-week-old C57BL/6J mice . Livers were isolated 4 weeks after injection and next generation sequencing showed an average of 1.4% and 5.4% precise prime editing with the low and high AAV doses, respectively (Figure 4D ). This demonstrates that PECO-Mini can be efficiently delivered by dual AAVs for in vivo prime editing.

Serum testosterone dropped 60% in treated stallions at 12 h post-injection. Testes were collected 12 h post-injection and assessed for differential gene expression. Project description:We isolated whole livers from male TDG knockout livers (4-month post tamoxifen injection) and HCC, along with their age matched male control livers. RNA was extracted from the samples as per RNAzol manufacturer guidelines with DNase treatment. RNA was then analyzed by bioanalyzer and subjected to sequencing using the Illumina NextSeq 500 platform.

Project description:Adult male Sprague Dawley-derived rats were housed and fed as described previously (Dail et al., 2007). Three rats were treated i.p. with PB in saline at 80 mg/kg body weight/day (treated) for 5 days as described previously (Ma and Chambers; 1995). Following sacrifice on day 5, the livers were removed and stored at -80 C as previously described (Dail et al., 2007). Five 8 µm sections were taken from each of the three control and three experimental livers resulting in fifteen sections from each group using a Triangle Biomedical Supply Co Minotome Plus (Durham, NC) microtome. Differential detergent fractionation two dimensional liquid chromatography electrospray ionization tandem mass spectrometry (DDF 2D LC ESI MS2) and systems biology modeling were used to identify alterations in toxicologically relevant hepatic molecular functions and biological processes in the livers of rats
