Project description:The role of PGC1alpha in breast cancer lung metastasis is largely unknown. We used expression data from lung metastatic explants overexpressing PGC1alpha or control, treated with phenformin to understand global gene expression changes which occur in a PGC1alpha context and under phenformin treatment. We used expression data to understand the pathways and genes that may lead to lung metastasis in a PGC1alpha overexpression setting.

Project description:To understand global expression changes in a knockdown of PGC1alpha (siPGC1alpha) vs control (siControl) in a lung metastatic cell line (4175) Metabolic adaptations play a key role in fuelling tumour growth. However, less is known regarding the metabolic changes that promote cancer progression to metastatic disease. Herein, we reveal that PGC-1a expression and activity are differentially regulated depending on breast cancer metastatic sites. Breast cancer cells that preferentially metastasize to lung display striking upregulation of PGC-1a expression. PGC-1a promotes breast cancer cell migration and invasion in vitro and augments lung metastasis in vivo. MDA-MB-231 lung variant (4175) cells were treated with a short interfering RNA (siRNA) for human PGC-1a (PPARG1A) or control siRNA for 48hrs 24 hours post-plating cells.

Project description:We investigated the molecular mechanisms for osteolytic bone metastasis by selecting human lung cancer cell line subpopulations with elevated metastatic activity and validating genes that are overexpressed in these cells. A bone-seeking squamous lung cancer cell line (HARA-B4) was established by sequentially injecting parental HARA cells into the left ventricle of male 5-week-old nude mice 4 times. The parental line HARA and highly bone-metastatic line HARA-B4 were analized. Total of two samples. No replicates.

Project description:Osteosarcoma (OS) is the malignant bone tumor with a high tendency to metastasize to the lung, where the molecular mechanisms are unclear. The mouse OS cell line LM8 has been isolated originally from the Dunn OS cell line by in vivo selection as a subline with a high metastatic potential to the lung. We used gene chip-based global gene expression analysis of differential screening between parental Dunn and LM8 cells in order to reveal genes predominantly expressed in LM8 cells, which correlate with high metastatic potential.

Project description:Widespread metastasis is the major cause of human lung cancer-related deaths, but there is much to be elucidated about underlying mechanism. Based on the genome-wide expression analysis of the highly metastatic lung cancer cell line NCI-H460-LNM35 (LNM35), we identified a cancer metastasis signature composed of 45 genes with previously uncharacterized features but well-known associations with human cancers including CXCL1, IER3, PTTG1, DAD1, METAP2, E-cadherin and galectin 3. In addition, the 45-gene metastasis signature gene set was significantly associated with a subset of primary tumors with poor prognosis not only in the lung but also the breast.
