Project description:HuR shRNA adenoviruses were delivered into WT or humanized mice intravenously at 2 × 109 pfu/mouse for both control virus and HuR shRNA virus . After seven days, liver tissue samples were harvested after a four hours fasting and stored immediately in liquid nitrogen till further analysis.The frozen liver tissue samples were homogenized in Trizol reagent (Invitrogen) using TissueLyser LT system (Qiagen). The isolated RNA was purified by MagMAX RNA Extraction Kit (ThermoFisher) and the construction of strand specific sequencing libraries using TruSeq Stranded Total RNA Prep kit (Illumina) and the sequencing was performed at NHLBI DNA Sequencing and Genomics Core using Illumina HiSeq 3000 paired-end sequencing platform.
Project description:Genome wide DNA methylation profiling of liver tissues from 17 months old male mice belonging to either the C57BL/6J, the CD1 or the Sv129Ev strain and exposed to LCPS. The Illumina Infinium Mouse Methylation Beadchip was used to obtain DNA methylation profiles across approximately 285,000 CpGs in liver samples. Samples included 19 C57Bl/6J mice, 12 CD1 mice, and 16 Sv129Ev mice.
Project description:Mice wild type or knocked-out for the MyD88 gene specifically in liver, were recruited for this expression profiling experiment. Each group of mice (WT versus LKO) were fed with a control diet or a high fat diet. Then mice were sacrificed and liver samples form were processed for RNA extraction. Total liver RNA of each sample was then pooled with those of the same group and treatment for microarray hybridization.
