Project description:Immunodeficient mouse models have been valuable for studies of human hematopoiesis, but high-fidelity recapitulation of erythropoiesis in most xenograft recipients remains elusive. Recently developed immunodeficient and Kit mutant mice, however, have provided a suitable background to achieve higher-level human erythropoiesis after long-term hematopoietic engraftment. While there has been some characterization of human erythropoiesis in these models, a comprehensive analysis of various developmental stages has not yet been reported. Here, we have utilized cell surface phenotypes, morphologic analyses, and molecular studies to fully characterize human erythropoiesis from multiple developmental stages in immunodeficient and Kit mutant mouse models following long-term hematopoietic stem and progenitor cell engraftment. We show that human erythropoiesis in such models demonstrates complete maturation and enucleation, as well as developmentally appropriate globin gene expression. These results provide a framework for future studies to utilize this model system for interrogating disorders affecting human erythropoiesis and for developing improved therapeutic approaches.

Project description:Investigating the intra-lung human immune responses to SARS-CoV-2, and how immune signatures temporally correlate to distinct histopathological manifestations of disease in the human lung, is not possible in human patients. While current non-human primate and rodent models have proven instrumental for evaluating COVID-19 treatments and vaccines, their non-human nature precludes faithful recapitulation of fundamental host-pathogen interactions and immunopathogenesis, particularly those governing severe COVID-19 disease. Here, using different mouse models engrafted with human lung tissues and human immune hematopoietic cells, we provide a unique picture of how differential human hematopoietic reconstitution, and immune signatures upon infection, correlate with distinct histopathology during SARS-CoV-2 infection. To do so, we engrafted pairs of human fetal lung xenografts (fLX) harboring lung-resident hematopoietic lineages into immunodeficient NRG mice (NRG-L), or into NRG-Flk2-/-Flt3LG mice co-engrafted with components of a human immune system (HIS-NRGF-L). Strikingly, while SARS-CoV-2 inoculation of NRG-L mice resulted in productive infection and extensive histopathological features typically observed in the lungs of severe COVID-19 patients, HIS-NRGF-L were able to rapidly control infection while preserving tissue integrity. Distinct histopathological outcomes were governed by differential proteomics and phospho-proteomics signatures and underscored the USP18-ISG15 pathway as a top immunomodulatory pathway defining effective control of viral replication and maintenance of tissue integrity in human lung tissue during SARS-CoV-2 infection. Our work highlights fLX-engrafted mice as a powerful platform to investigate the molecular basis that drives effective immune control of SARS-CoV-2, and open avenues for the development of innovative immunotherapies targeting the USP18-ISG15 pathway to alleviate severe COVID-19.

Anemia and blood shortages has become global challenges, with current finite pharmacological options like EPO or glucocorticoids having limitations, especially in genetic anemias like Diamond-Blackfan anemia (DBA), calling for novel candidates in stimulating the erythropoiesis. We designed the primary human hematopoietic stem and progenitor cells (HSPCs) chemicals screening system in erythropoiesis and unexpectedly discovered a BRAF inhibitor, effectively against BRAFV600E mutant cancers, delaying erythroid differentiation while promoting progenitors’ proliferation. Further research demonstrated its efficacy in cytokine-restricted conditions and in samples from erythroid disorder patients. Mechanistically, BRAF inhibitors paradoxically activated the RAF-MEK-ERK/MAPK pathway. Unlike oncogenic BRAFV600E impaired hematopoiesis and erythropoiesis via AP-1 hyperactivation, BRAF inhibitors minimally affected HSPCs self-renewal and differentiation. In vivo studies further exhibited BRAF inhibitors' potential to enhance human hematopoiesis and erythropoiesis in severe immunodeficient mouse models and alleviate anemia symptoms in Rpl11 haploinsufficiency DBA models and other anemia models. Project description:Erythropoiesis is a crucial process in hematopoiesis, yet it remains highly susceptible to disruption by various diseases, which significantly contribute to the global challenges of anemia and blood shortages. Current treatments like erythropoietin (EPO) or glucocorticoids often fall short, especially for hereditary anemias such as Diamond-Blackfan anemia (DBA). To uncover new erythropoiesis-stimulating agents, we devised a screening system using primary human hematopoietic stem and progenitor cells (HSPCs). We discovered that BRAF inhibitors (BRAFi), commonly used to treat BRAFV600E melanoma, can unexpectedly and effectively promote progenitor cell proliferation by temporarily delaying erythroid differentiation. Notably, these inhibitors exhibited pronounced efficacy even under cytokine-restricted conditions and in patient samples of DBA. Mechanistically, although these BRAFi inhibit the MAPK cascade in BRAFV600E mutant cells, they paradoxically act as amplifiers in wild-type BRAF cells, potently enhancing the cascade. Furthermore, we found that while the oncogenic BRAFV600E mutation disrupts hematopoiesis and erythropoiesis through AP-1 hyperactivation, BRAFi minimally impact HSPC self-renewal and differentiation. In vivo studies have shown that BRAFi can enhance human hematopoiesis and erythropoiesis in severe immunodeficient mouse models and alleviate anemia in the Rpl11 haploinsufficiency DBA model, as well as other relevant anemia models. This discovery underscores the role of the MAPK pathway in hematopoiesis and positions BRAFi as a promising therapeutic option for improving hematopoietic reconstitution and treating anemias, including DBA.
