Proteogenomic characterization of microbial communities fueled by metal-sulfide oxidation of inactive hydrothermal chimneys

Project description:Iron-sulfur minerals such as pyrite are found in many marine benthic habitats. At deep-sea hydrothermal vent sites they occur as massive sulfide chimneys. Hydrothermal chimneys formed by mineral precipitation from reduced vent fluids upon mixing with cold oxygenated sea water. While microorganisms inhabiting actively venting chimneys and utilizing reduced compounds dissolved in the fluids for energy generation are well studied, only little is known about the microorganisms inhabiting inactive sulfide chimneys. We performed a comprehensive meta-proteogenomic analysis combined with radiometric dating to investigate the diversity and function of microbial communities found on inactive sulfide chimneys of different ages from the Manus Basin (SW Pacific). Our study sheds light on potential lifestyles and ecological niches of yet poorly described bacterial clades dominating inactive chimney communities.

Project description:To assess the risk that mining of seafloor massive sulfides (SMS) from extinct hydrothermal vent environments has for changing the ecosystem irreversibly, we sampled SMS analogous habitats from the Kairei and the Pelagia vent fields along the Indian Ridge. In total 19.8 million 16S rRNA tags from 14 different sites were analyzed and the microbial communities were compared with each other and with publicly available data sets from other marine environments. The chimneys appear to provide habitats for microorganisms that are not found or only detectable in very low numbers in other marine habitats. The chimneys also host rare organisms and may function as a vital part of the ocean's seed bank. Many of the reads from active and inactive chimney samples were clustered into OTUs, with low or no resemblance to known species. Since we are unaware of the chemical reactions catalyzed by these unknown organisms, the impact of this diversity loss and bio-geo-coupling is hard to predict. Given that chimney structures can be considered SMS analogues, removal of sulfide deposits from the seafloor in the Kairei and Pelagia fields will most likely alter microbial compositions and affect element cycling in the benthic regions and probably beyond.

Key Factors Influencing Rates of Heterotrophic Sulfate Reduction in Active Seafloor Hydrothermal Massive Sulfide Deposits.

Project description:Hydrothermal vents are thermally and geochemically dynamic habitats, and the organisms therein are subject to steep gradients in temperature and chemistry. To date, the influence of these environmental dynamics on microbial sulfate reduction has not been well constrained. Here, via multivariate experiments, we evaluate the effects of key environmental variables (temperature, pH, H2S, [Formula: see text], DOC) on sulfate reduction rates and metabolic energy yields in material recovered from a hydrothermal flange from the Grotto edifice in the Main Endeavor Field, Juan de Fuca Ridge. Sulfate reduction was measured in batch reactions across a range of physico-chemical conditions. Temperature and pH were the strongest stimuli, and maximum sulfate reduction rates were observed at 50°C and pH 6, suggesting that the in situ community of sulfate-reducing organisms in Grotto flanges may be most active in a slightly acidic and moderate thermal/chemical regime. At pH 4, sulfate reduction rates increased with sulfide concentrations most likely due to the mitigation of metal toxicity. While substrate concentrations also influenced sulfate reduction rates, energy-rich conditions muted the effect of metabolic energetics on sulfate reduction rates. We posit that variability in sulfate reduction rates reflect the response of the active microbial consortia to environmental constraints on in situ microbial physiology, toxicity, and the type and extent of energy limitation. These experiments help to constrain models of the spatial contribution of heterotrophic sulfate reduction within the complex gradients inherent to seafloor hydrothermal deposits.

Massive seafloor mounds depict potential for seafloor mineral deposits in the Great South Basin (GSB) offshore New Zealand.

Project description:Seafloor mounds are enigmatic features along many continental margins and are often interpreted as gas hydrate pingoes, seep deposits, mud volcanoes, or hydrothermal mounds. When such mounds occur in basins with past volcanic activities, they have the potential to host seafloor metalliferous deposits, which is generally overlooked. Using geophysical datasets, we document the fluid plumbing systems that promoted the formation of seafloor mounds in the Great South Basin (GSB), offshore New Zealand. We also investigate these mounds as potential seafloor metalliferous deposits. Our results reveal 9 seafloor mounds (~ 137 m high) above gigantic (~ 5.4 km high) fluid escape pipes that are associated with deeper crystalline rocks. The structural make-up of the mounds, their geospatial relationships with the pipes and intrusive rocks, and geophysical properties suggest a primary volcanic or hydrothermal origin for the culpable fluids and mounds respectively. Fluids derived from deeper coal beds and shallow foraminiferal oozes in the basin constitute secondary fluid sources focused along polygonal faults and lateral flow cells. A main sub-vertical and minor lateral fluid plumbing patterns are proposed. The relationship between the mounds, pipes, underlying intrusive rocks, and upward routing of mineral-rich fluids could have implications for the formation of ore-grade mineral deposits on the seafloor in the GSB.
