Project description:To know BRs was how to cause transgenic line L100 leaves changed, we took the sixth leaves of transgenic line L100 to RNA-seq sequence. Total RNA was extracted from the sixth leaves of six-leaf stage of transgenic line L100 and wild type Q319. cDNA library was builded in the BIOMARKER, 100.20M original reads were sequenced based on Illumina HiSeq2000, and through rRNA, low quality fragment DNA of filtering, ultimately, we obtained 69.01M high quality clean reads. Compared with reference gene sequence(AGPv3), according to genes function annotation, we found 201 different expression genes. We classified the different expression genes according to Cellular Component, Molecular Function and Biological Process. It showed that BRs could affect many genes expression involved in different biological process.

Leaf margins of indeterminate Arabidopsis leaves

Project description:Leaves are flat determinate organs derived from indeterminate shoot apical meristems. The presence of a specific leaf meristem is debated, as anatomical features typical of meristems are not present in leaves. Here we demonstrate that multiple NGATHA (NGA) and CINCINNATA-class-TCP (CIN-TCP) transcription factors act redundantly to suppress activity of a leaf margin meristem in Arabidopsis thaliana, and that their absence confers persistent marginal growth of leaves, cotyledons and floral organs. The marginal meristem is activated by the juxtaposition of adaxial and abaxial domains and maintained by WOX homeobox transcription factors, but other margin elaboration genes are dispensable for its maintenance. This genetic framework parallels the morphogenetic program of shoot apical meristems and may represent a relic from an ancestral shoot system from which seed plant leaves evolved.

RNA-seq analysis of dark-induced senescence of the leaves of Ran-GDP and Ran-GTP overexpression transgenic plants

Project description:Comparing transcriptome of the leaves of Ran-GDP and Ran-GTP overexpression transgenic plants before and after dark incubation, we discovered 851 differentially expressed genes that designated as Ran-regulated genes. The RNA-seq result confirmed the early senescence phenotype of Ran-GTP overexpression plants and the up-regulation of several signaling pathways, implied their positive roles in Ran-regulated senescence and suggest potential downstream targets of Ran. This study provided a mechanism that senescence process could be regulated by nuclear transportation machinery in Arabidopsis.

Transcriptome profile for leaves of rice plants subjected to severe reproductive stage drought stress

Project description:To understand how the qDTY12.1 interacts with other genes within the genome to give better physiological adaptations and better yield; the leaves transcriptome of DTY12.1 parents (Vandana , Way Rarem) and DTY12.1 NIL (481-B) were compared under control and drought conditions. Drought induced gene expression was studied in the leaves of the rice plants subjected to severe reproductive stage drought. Seeds of Vandana, Way Rarem, and 481-B were sown into rotovated soil at a rate of 2.0 g m-1 into plots of 3 rows X 3 m. The three genotypes were sown in three replications in a randomized complete block design.

Expression data of leaves from transgenic barley expressing wheat Lr34 gene

Project description:The wheat gene Lr34 (Yr18/Pm38/Sr57/Ltn1) encodes a putative ABCG-type of transporter and is a unique source of disease resistance providing durable and partial resistance against multiple fungal pathogens. Lr34 has been found to be functional as a transgene in barley. We used microarrays to decipher the changes in global gene expression governed by Lr34 expression in barley. Leaves from two independent lines of transgenic barley plants were selected at seedling (two weeks old plants) and mature (six weeks old plants) stages of development for RNA extraction and hybridization on Affymetrix microarrays. We used three biological replicates for expression profiles. Wild type plants served as controls.
