Project description:Nitrogen starvation is an efficient environmental pressure used to increase lipid accumulation and oil droplet formation in microalgal cells. Various studies focused on metabolic changes occurring in microalgae in nitrogen starvation conditions, but the mechanisms at the basis of these changes are not completely understood. Between microalgae, green algae, with more than 7000 species growing in a variety of habitats, have been frequently studied for energy purposes, but also as source of bioactive extracts/compounds. In this study, de novo transcriptome of the green algae Tetraselmis suecica has been performed in order to (1) deeply study its response to nitrogen starvation, (2) to look for enzymes with antioxidant capacity and for polyketide synthases (PKSs), (3) if present, to evaluate if nutrient starvation can influence their expression levels.

New molecular insights on the response of the green alga Tetraselmis suecica to nitrogen starvation.

Project description:Microalgae are currently considered one of the most promising resources for biofuel production, aquaculture feedstock and new pharmaceuticals. Among them, green algae of the genus Tetraselmis are extensively studied for their lipid accumulation in nutrient-starvation conditions. In this paper, we present the full-transcriptome of Tetraselmis suecica and differential expression analysis between nitrogen-starved and -repleted conditions (at stationary phase) focusing not only on lipid metabolism but giving new insights on nutrient starvation responses. Transcripts involved in signal transduction pathways, stress and antioxidant responses and solute transport were strongly up-regulated when T. suecica was cultured under nitrogen starvation. On the contrary, transcripts involved in amino acid synthesis, degradation of sugars, secondary metabolite synthesis, as well as photosynthetic activity were down-regulated under the same conditions. Among differentially expressed transcripts, a polyketide synthase and three lipoxygenases (involved in the synthesis of secondary metabolites with antipredator, anticancer and anti-infective activities) were identified, suggesting the potential synthesis of bioactive compounds by this microalga. In addition, the transcript for a putative nitrilase, enzyme used in nitrile bioremediation, is here reported for the first time for T. suecica. These findings give new insights on T. suecica responses to nutrient starvation and on possible biotechnological applications for green algae.

Techno-Functional Properties of Crude Extracts from the Green Microalga Tetraselmis suecica.

Project description:A mild fractionation process to extract functional biomolecules from green microalgae was implemented. The process includes bead milling, centrifugation, and filtration with several membrane cut-offs. For each fraction, the corresponding composition was measured, and the surface activity and gelation behavior were determined. A maximum protein yield of 12% was obtained in the supernatant after bead milling and between 3.2 and 11.7% after filtration. Compared to whey protein isolate, most of the algae fractions exhibited comparable or enhanced functionality. Surface activity for air-water and oil-water interfaces and gelation activities were notably superior for the retentate fractions compared to the permeates. It is proposed that such functionality in the retentates is due to the presence of hydrophobic compounds and molecular complexes exhibiting a similar behavior as Pickering particles. We demonstrated that excellent functionality can be obtained with crude fractions, requiring minimum processing and, thus, constituting an interesting option for commercial applications.
