Project description:Estrogen (E2)-dependent gene regulation mediated by estrogen receptor alpha (ERα) plays a mitogenic role in ER-positive breast cancer cells. Although clinical applications of selective estrogen receptor modulators (SERMs), which directly interact with ERα to alter ERα activity, have been effective as a first line of treatment for breast cancer patients, a large subset of the patients will develop resistance after prolonged use of SERMs. Thus, there is a great need to develop alternative therapeutic strategies for SERM-resistant breast cancers. Here, we describe the potential use of the bromodomain family member protein (BRD) selective bromodomain inhibitor, JQ1, to alter E2-dependent gene expression program and inhibit E2-dependent growth of breast cancer cells. We show that each family member has partially redundant roles as ERα coregulators that are required for ERα-mediated gene transcription. Furthermore, we demonstrate the function of BRD3 as a molecular sensor of total BRD activity by the compensatory control of its protein levels. In addition, BRD3 colocalizes with a subset of ERα binding sites (ERBSs) that are enriched for active enhancer features and associated with highly E2-induced genes. Collectively, we illustrate a critical role of the BET family members in ERα dependent gene expression.

Functional coordination of BET family proteins

Project description:Bromodomain and extraterminal proteins (BET) are epigenetic adaptors that play critical roles in gene regulation. This protein family contains two conserved bromodomains which can mediate the interaction with acetylated histone lysine residues and/or other acetylated proteins, and one extraterminal (ET) domain. BET protein inhibition has been recognized as a promising therapeutic strategy for several diseases, including acute myeloid leukaemia, castration resistant prostate cancer, and triple negative breast cancer. BET inhibition by a specific inhibitor JQ1 caused an impairment in various brain functions but the underlying molecular mechanisms are not well understood. Among BET family members, BRD4 has been recognized for its prominent role in epigenetic regulation of gene expression. However, we have found an extensive functional redundancy and coordination among all three major BET family members (BRD2/3/4) during the activity-dependent gene induction in neurons, which is also reflected in learning and memory behavior. Our study further revealed the molecular hierarchy in the coordination of BET family proteins dissecting their distinctive functions during gene induction required for proper brain function and plasticity. Understanding the functional coordination of BET family proteins at the molecular level would be beneficial to the development of more selective interventions for various diseases that are caused by dysregulated epigenetic pathways.

BET family members Bdf1/2 modulate global transcription initiation and elongation in Saccharomyces cerevisiae (ChEC-Seq)

Project description:Human BET family members are promising targets in the therapy of cancer and immunoinflammatory diseases, but their mechanism of action and functional redundancies are poorly understood. Yeast BET factors Bdf1/2 were previously proposed to act as anchors for coactivator TFIID. We investigated their genome wide roles in transcription and found that, while they cooperate with TFIID at many genes, their contributions to transcription are often significantly different. Bdf1/2 co-occupy the majority of yeast promoters and affect preinitiation complex formation by participating in recruitment of TFIID, Mediator and basal factors to chromatin. Surprisingly, we discovered that hypersensitivity of genes to Bdf1/2 depletion results from combined defects in initiation of transcription and early elongation. Bdf1/2 are critical components of yeast transcriptional machinery with many functional similarities to human BET proteins, most notably Brd4.

Chromatin profiling in human neurons reveals aberrant roles for histone acetylation and BET family proteins in schizophrenia

Project description:Schizophrenia (SZ) is a psychiatric disorder with complex genetic risk dictated by interactions between hundreds of risk variants. Epigenetic factors, such as histone posttranslational modifications (PTMs), have been shown to play critical roles in many neurodevelopmental processes, and when perturbed may also contribute to the precipitation of disease. Here, we apply an unbiased proteomics approach to evaluate combinatorial histone PTMs in human induced pluripotent stem cell (hiPSC)-derived forebrain neurons from individuals with SZ. We observe hyper-acetylation of H2A.Z and H4 in neurons derived from SZ cases, results that were confirmed in postmortem human brain. We demonstrate that the bromodomain and extraterminal (BET) protein, BRD4, is a bona fide ‘reader’ of H2A.Z acetylation, and further provide evidence that BET family protein inhibition ameliorates transcriptional abnormalities in patient-derived neurons. Thus, treatments aimed at alleviating BET protein interactions with hyperacetylated histones may aid in the prevention or treatment of SZ.
