Project description:Transcriptional control of any biological process occurs through the action of one to many gene regulatory networks (GRNs), but studies analyzing GRN interaction are lacking. To address this, we tested for the first time in vivo the hypothesis that two independent GRNs, which are active during formation of two discrete skeletal cell types (immature chondrocytes and osteoblasts), interact during differentiation of a third skeletal cell type (mature chondrocytes). These three cell types were isolated from the mouse embryo specifically using laser capture microdissection, and then RNA was extracted. Multiple analyses of corresponding RNA-seq data supported the hypothesis. Gene co-expression network analyses of differentially expressed genes suggested that one GRN containing the chondrogenic transcription factor Sox9 characterizes immature chondrocytes, one GRN containing the osteogenic transcription factor Runx2 characterizes osteoblasts, and both GRNs operate in mature chondrocytes. Indeed, mature chondrocytes differentially expressed fewer genes than the other cell types, consistent with the idea that overlapping actions of the immature chondrocyte and osteoblast GRNs regulate mature chondrocytes. Clustering analyses provided molecular insights into potential GRN interactions. Several genes in mature chondrocytes had expression levels that represented an averaging between levels in the immature chondrocyte and osteoblast. Interestingly, expression levels of one gene cluster, containing the hallmark mature chondrocyte genes Collagen type 10a1 and Indian hedgehog, suggested a synergistic interaction between immature chondrocyte and osteoblast GRNs. In addition to identifying novel genes expressed in mature chondrocytes, these results outline a novel in vivo experimental system through which to understand GRN organization and interaction.

Project description:Joint diseases are often characterized by inflammatory processes resulting in pathological changes in joint tissues, including cartilage degradation and release of components to the synovial fluid. The complement system plays a central role in promoting the inflammation. Since several cartilage proteins are known to interact with complement, causing either activation or inhibition of the system, we aimed to investigate these interactions comprehensively. Bovine cartilage explants were cultured with interleukin-1alpha (IL-1a) to induce cartilage degradation, followed by incubation with human serum to allow interactions with complement. Label-free selected reaction monitoring (SRM) mass spectrometry (MS) was then used to specifically quantify complement proteins interacting with the cartilage explant. In parallel, the time-dependent degradation of cartilage was detected using tandem MS (MS/MS). Complement proteins resulting from activation of the classical and alternative pathway as well as the terminal pathway were detected on IL-1a stimulated cartilage at time points when clear alterations in the extracellular matrix composition had occurred. To confirm SRM results indicating complement activation, increased levels of the complement activation product C4d were detected by ELISA in serum after incubation with IL-1a stimulated cartilage. Further, typical activated (cleaved) C3 fragments were detected by western blotting of urea extracts of IL-1a stimulated cartilage. No complement activation was triggered by cartilage cultured in the absence of IL-1a. Components released from IL-1a stimulated cartilage during culture had an inhibitory effect on complement activation. These were released after a longer incubation period with IL-1a and may represent a feedback reaction to cartilage-triggered complement activation observed after a shorter incubation period.

Project description:Successfully replacing damaged cartilage with tissue-engineered constructs requires integration with the host tissue and could benefit from leveraging the native tissue's intrinsic healing capacity; however, efforts are limited by a poor understanding of how cartilage repairs minor defects. Here, we investigated the conditions that foster natural cartilage tissue repair to identify strategies that might be exploited to enhance the integration of engineered/ grafted cartilage with host tissue. We damaged porcine articular cartilage explants and using a combination of pulsed SILAC-based proteomics, ultrastructural imaging, and catabolic enzyme blocking strategies reveal that integration of damaged cartilage surfaces is not driven by neo-matrix synthesis, but rather local depletion of proteoglycans. ADAMTS4 expression and activity are upregulated in injured cartilage explants, but integration could be reduced by inhibiting metalloproteinase activity with TIMP3. These observations suggest that catabolic enzyme-mediated proteoglycan depletion likely allows existing collagen fibrils to undergo cross-linking, fibrillogenesis, or entanglement, driving integration. Catabolic enzymes are often considered pathophysiological markers of osteoarthritis. Our findings suggest that damage-induced upregulation of metalloproteinase activity may be a part of a healing response that tips towards tissue destruction under pathological conditions and in osteoarthritis, but could also be harnessed in tissue engineering strategies to mediate repair. Statement of significance: Cartilage tissue engineering strategies require graft integration with the surrounding tissue; however, how the native tissue repairs minor injuries is poorly understood. We applied pulsed SILACbased proteomics, ultrastructural imaging, and catabolic enzyme blocking strategies to a porcine cartilage explant model and found that integration of damaged cartilage surfaces is driven by catabolic enzyme-mediated local depletion of proteoglycans. Although catabolic enzymes have been implicated in cartilage destruction in osteoarthritis, our findings suggest that damage-induced upregulation of metalloproteinase activity may be a part of a healing response that tips towards tissue destruction under pathological conditions.
