Project description:Objective: Physical exercise and vitamin E are considered effective treatments of nonalcoholic fatty liver and other metabolic diseases. However, vitamin E has also been shown to interfere with the adaptation to exercise training, in particular for the skeletal muscle. Here, we studied the hypothesis that vitamin E also interferes with the metabolic adaptation of the liver to acute exercise. Methods: Male C57BL6/N mice were fed either control or vitamin E (α-tocopherol)- supplemented antioxidant diet during four weeks before being subjected to a non-exhaustive treadmill run. We assessed the acute transcriptional response of the liver as well as plasma corticosterone and free fatty acid (FFA) concentrations and monitored respiration in the exercising mice. Results: Vitamin E counteracted exercise-induced hepatic inflammation and altered the metabolic adaptation of the liver on the level of gene transcription. Vitamin E interfered with the upregulation of key metabolic regulators and caused a paradoxical increase in transcripts involved in lipid and cholesterol synthesis, processes negatively regulated by FFA and usually suspended during physical exercise. Whole-body energy consumption as well as corticosterone levels and signalling were similar, arguing against acute differences in fatty acid oxidation or glucocorticoid action. Conclusions: Our results show that vitamin E supplementation alters the inflammatory and transcriptional response of the liver to physical exercise. These effects of vitamin E could, on the long run, result in an impaired adaptation of the liver to physical exercise. Since exercise is clearly beneficial for general health and in reducing inflammation and dyslipidemia in nonalcoholic fatty liver disease, the interference of vitamin E with these processes may speak for a cautious use as dietary supplement.

There is growing evidence that reactive oxygen species (ROS) are necessary for some of these adaptations and antioxidants may be used to investigate this effect. This study aimed to determine the effects of exercise and/or antioxidant supplementation on myocardial and vascular endothelium gene expression. Male Wistar rats were divided into four groups: i) endurance exercise (90min of treadmill running 4d/week, 14 weeks); ii) antioxidant-treated; iii) antioxidant and endurance exercise and iv) control. The supplemented animals received Vitamin E (1000 IU/kg diet) and ï¡-lipoic acid (1.6 g/kg diet) mixed with rat chow. cDNA microarray analysis was performed using purified endothelial RNA from myocardial and coronary artery endothelial cells and showed that the expression levels of 35, 40 and 40 genes were altered for groups i, ii, and iii respectively compared to control. Differentially expressed genes were analysed using the KEGG pathway database, hierarchical cluster and DAVID analysis. These analyses revealed that a gene involved in cardiovascular disease progression, Ras homolog gene family member A (RhoA) was down-regulated by exercise, upregulated by antioxidant supplementation and the combination of exercise and antioxidant blunted both effects. These findings were confirmed by real-time PCR. Project description:Physical exercise is beneficial for general health and is an effective treatment for metabolic disorders. Vitamin E is widely used as dietary supplement and is considered to improve non-alcoholic fatty liver disease by reducing inflammation and dyslipidemia. However, increased vitamin E intake may interfere with adaptation to exercise training. Here, we explored how vitamin E alters the acute exercise response of the liver, an organ that plays an essential metabolic role during physical activity. Mice fed a control or an α-tocopherol-enriched diet were subjected to a non-exhaustive treadmill run. We assessed the acute transcriptional response of the liver as well as glucocorticoid signalling and plasma free fatty acids (FFA) and performed indirect calorimetry. Vitamin E interfered with the exercise-induced increase in FFA and upregulation of hepatic metabolic regulators, and it shifted the transcriptional profile of exercised mice towards lipid and cholesterol synthesis while reducing inflammation. Energy utilization, as well as corticosterone levels and signalling were similar, arguing against acute differences in substrate oxidation or glucocorticoid action. Our results show that high-dose vitamin E alters the metabolic and inflammatory response of the liver to physical exercise. The interference with these processes may suggest a cautious use of vitamin E as dietary supplement.
