RNA-seq analysis in mouse cortical extracts with deletion of STI1 TPR1 domain

Project description:Long RNA-sequencing was performed on 5 cortex samples from STI1 wt/wt and 5 STI1 dTPR/dTPR mice. RIN values for these samples ranged between 8.2 and 8.7. Illumina sequencing yielded an average of 797,963 reads per sample, with 93.69% reads mapping rate to the mouse genome. We first verified the complete absence of STI1 exons 2 and 3 in the dTPR/dTPR samples compared to the wt/wt control tissues where these axons were both expressed. Principal component analysis and sample distance matrix analyses did not segregate between the two groups, indicating minimal differences between them. Furthermore, thorough RNA sequencing analysis did not reveal any significant changes in transcripts passing FDR correction (padj<0.05). Briefly, there were 85 gene transcripts (representing 18 unique genes) that passed p.value<0.05 and whose levels were changed by more than 1.5 fold increase (or 0.75 decrease)

Project description:[original Title] Expression profiling of mouse embryonic fibroblasts with or without a deletion in the helicase domain of the Werner Syndrome gene homologue treated with hydrogen peroxide Werner syndrome (WS) is a rare disorder characterized by the premature onset of a number of age-related diseases. The gene responsible for WS is believed to be involved in different aspects of transcription, replication, and/or DNA repair. In addition to genomic instability, WS cells exhibit oxidative stress. In this report, we have examined the impact of exogenous peroxide on the expression profile of mouse embryonic fibroblasts lacking part of the helicase domain of the WRN homologue. This expression profile was compared to hydrogen peroxide treated wild type mouse embryonic fibroblasts. Interestingly, untreated mouse embryonic fibroblasts with a deletion of the helicase domain of Wrn already exhibited down regulation of several biological processes decreased by hydrogen peroxide in wild type cells.

DOT1L deletion impairs the development of cortical Parvalbumin-expressing interneurons

Project description:Our group has reported that the histone methyltransferase DOT1L is necessary for proper cortical plate development and layer distribution of glutamatergic neurons, however, its specific role on cortical interneuron development has not yet been explored. Here, we demonstrate that DOT1L affects interneuron development in a cell-autonomous manner. Deletion of Dot1l in MGE-derived interneuron precursor cells results in an overall reduction and altered distribution of GABAergic interneurons in the cortical plate at postnatal day (P) 0. Furthermore, we observed an altered proportion of GABAergic interneurons in the cortex and striatum at P21 with a significant decrease in Parvalbumin (PVALB)-expressing interneurons. Altogether, our results indicate that reduced numbers of cortical interneurons upon DOT1L deletion results from altered postmitotic differentiation/maturation.
