We find miR319 is involved in citrus adapation to long-term boron toxicity via targeting a MYB gene, Ciclev10000756m.g.v1.0, which is homologus with several MYBs that modulate lateral root development in Arabidopsis.

Project description:Our previous studies indicated that long-term B-toxicity was mainly limited to leaves of Citrus species; alternations of cell wall structure in vascular bundles were involved in tolerance to B-toxicity. Here, miRNAs and their expression pattern were first identified from B-treated Citrus sinensis (tolerant) and C. grandis (intolerant) leaves with high-throughput sequencing in order to identify miRNAs that might be involved in tolerance to B-toxicity. Results: 51 (23 conserved and 28 novel) miRNAs in C. grandis and 20 (6 conserved and 14 novel) in C. sinensis were differentially expressed after B-toxic treatment, respectively. Illumina sequencing results were validated by stem-loop qRT-PCR, and 82.5% qRT-PCR data coincided with those from direct sequencing. Among the differentially expressed miRNAs, miR395a and miR397a were the most significantly up-regulated in B-toxic C. grandis leaves, yet both were down-regulated in B-toxic C. sinensis ones. With modified 5’-RACE, four auxin response factor genes were confirmed as the real targets of miR160a, and two laccase (LAC) genes as those of miR397a, respectively. Localization of cell wall polysaccharides indicated that up-regulation of LAC4 resulted in secondary deposition of cell wall polysaccharides in regions near the pits of vessel elements in C. sinensis, and that down-regulation of both LAC17 and LAC4, via up-regulating their negative regulator miR397a, led to poorly developed vessel elements in C. grandis.Our findings demonstrated that miR397a played a pivotal role in woody Citrus tolerance to B-toxicity by targeting LAC17 and LAC4, of which both were responsible for secondary cell wall synthesis.

Project description:Boron (B) toxicity in Citrus is a common physiological disorder leading to reductions in both productivity and quality. Studies on how Citrus roots evade B toxicity may provide new insight into plant tolerance to B toxicity. Here, using Illumina sequencing, differentially expressed microRNAs (miRNAs) were identified in B toxicity-treated Citrus sinensis (tolerant) and C. grandis (intolerant) roots. The results showed that 37 miRNAs in C. grandis and 11 miRNAs in C. sinensis were differentially expressed when exposed to B toxicity. Among them, miR319, miR171, and miR396g-5p were confirmed via 5'-RACE and qRT-PCR to target a myeloblastosis (MYB) transcription factor gene, a SCARECROW-like protein gene, and a cation transporting ATPase gene, respectively. Maintenance of SCARECROW expression in B treated Citrus roots might fulfill stem cell maintenance, quiescent center, and endodermis specification, thus allowing regular root elongation under B-toxic stress. Down-regulation of MYB due to up-regulation of miR319 in B toxicity-treated C. grandis roots might decrease the number of root tips, thereby dramatically changing root system architecture. Our findings suggested that miR319 and miR171 play a pivotal role in Citrus adaptation to long-term B toxicity by targeting MYB and SCARECROW, respectively, both of which are responsible for root growth and development.

Seedlings of Al-tolerant Citrus sinensis and Al-intolerant Citrus grandis were fertigated with nutrient solution containing 0 and 1.0 mM AlCl3•6H2O. Thereafter, we investigated the Al-toxicity-induced alterations of transcriptomics in leaves by RNA-Seq. Results: Using RNA-seq, we isolated 1162 (181) up- and 496 (234) downregulated genes from Al-treated C. grandis (C. sinensis) leaves. Clearly, gene expression was less affected by Al-toxicity in C. sinensis leaves than in C. grandis ones. Several Al-toxicity-responsive genes homologous to known Al-tolerance genes: ALUMINUM SENSITIVE 3 (ALS3), multidrug and toxic compound extrusion (MATE), glutathione S-transferase (GST), L-galactose dehydrogenase（L-GalDH) and lipoxygenase (LOX) were identified in citrus leaves. Genes related to signal transduction, and sulfur transport and metabolism might also play a role in the higher Al-tolerance of C. sinensis. Conclusions: This is the first comparative investigation of transcriptomic responses in Al-treated citrus leaves. There were common and unique mechanisms for citrus Al-tolerance. These results provide a platform for further investigating the roles of genes possibly responsible for citrus Al-tolerance.

Project description:The mechanisms underlying tolerance to B-toxicity in plants are still controversial. Our previous studies indicated that B-toxicity is mainly limited to leaves in Citrus and that alternations of cell-wall structure in vascular bundles are involved in tolerance to B-toxicity. Project description:Self-germinated seedlings of Citrus sinensis and C. grandis were supplied with nutrient solution with 0 mM AlCl3·6H2O (control, -Al) or 1 mM AlCl3·6H2O (+Al) for 18 weeks. The DW (Dry weights) of leaf, stem, shoot and the whole plant of C. grandis were decreased and the ratio of root DW to shoot DW in C. grandis were increased by Al, whereas these parameters of C. sinensis were not changed by Al. Al treatment dramatically decreased the sulfur (S) content in C. grandis roots and the phosphorus (P) content in both C. sinensis and C. grandis roots. More Al was transported to shoots and leaves in C. grandis than in C. sinensis under Al treatment. Al treatment has more adverse effects on C. grandis than on C. sinensis, as revealed by the higher production of superoxide anion (O2 ·-), H2O2 and thiobarbituric acid reactive substace (TBARS) content in C. grandis roots. Via the Illumina sequencing technique, we successfully identified and quantified 12 and 16 differentially expressed miRNAs responding to Al stress in C. sinensis and C. grandis roots, respectively. The possible mechanism underlying different Al tolerance of C. sinensis and C. grandis were summarized as having following aspects: (a) enhancement of adventitious and lateral root development (miR160); (b) up-regulation of stress and signaling transduction related genes, such as SGT1, PLC and AAO (miR477, miR397 and miR398); (c) enhancement of citrate secretion (miR3627); (d) more flexible control of alternative glycolysis pathway and TCA cycle (miR3627 and miR482); (e) up-regulation of S-metabolism (miR172); (f) more flexible control of miRNA metabolism. For the first time, we showed that root development (miR160) and cell wall components (cas-miR5139, csi-miR12105) may play crucial roles in Al tolerance in citrus plants. In conclusion, our study provided a comprehensive profile of differentially expressed miRNAs in response to Al stress between two citrus plants differing in Al tolerance which further enriched our understanding of the molecular mechanism underlying Al tolerance in plants.
