Project description:Malaria causing parasites belonging to genus Plasmodium have a complex life cycle involving two hosts. Within these hosts parasites continuously shift between multiple morphologically distinct stages each of which has unique abilities to invade and survive in different host cell types. As repeatedly shown by independent studies, transcriptional profile of the parasite alters dramatically between stages which highlights the requirement for rapid and precise control of gene expression in these protozoans. However, genome wide studies revealed that plasmodium lacks orthologues of transcription factors found in other eukaryotes, has undefined promotor regions lacking conserved regulatory elements, has a lot smaller predicted specific TF to total proteome ratio compared to other eukaryotes. Instead, these parasites have been shown to contain a sophisticated histone modification machinery, stage specific control of mRNA decay rates, abundance of anti-sense RNA transcription and multiple epigenetic regulatory mechanisms . All these supporting evidences led to the general conception that plasmodium gene expression is primarily regulated by mechanisms other than TFs.

Project description:The acquisition of malaria immunity is both remarkably slow and unpredictable. At present, we know little about the malaria parasite genes that influence the host's ability to mount a protective immune response. Here, we show that a single-nucleotide polymorphism (SNP) resulting in a single amino acid change (S to F) in an ApiAP2 transcription factor in the rodent malaria parasite Plasmodium berghei (Pb) NK65 allowed infected mice to mount a T helper cell 1 (TH1)-type immune response that controlled subsequent infections. As compared to PbNK65S, PbNK65F parasites differentially expressed 46 genes, most of which are predicted to play roles in immune evasion. PbNK65F infections resulted in an early interferon-γ response and a later expansion of germinal centers, resulting in high levels of infected red blood cell-specific TH1-type immunoglobulin G2b (IgG2b) and IgG2c antibodies. Thus, the Pb ApiAP2 transcription factor functions as a critical parasite virulence factor in malaria infections.

Project description:The family of apicomplexa-specific proteins with DNA binding AP2 domains (ApiAP2s) includes sequence-specific transcription factors that are key regulators of development in malaria parasites. However, functions for the majority of ApiAP2 genes remain unknown. Here, a systematic knockout screen in Plasmodium berghei identifies ten ApiAP2 genes essential for mosquito transmission, of which four are critical for the formation of infectious ookinetes and three for sporogony. We describe unexpected non-essential functions for AP2-O and AP2-SP proteins in blood stages and identify AP2-G2 as a universal repressor active in both, asexual and sexual stages. Comparative transcriptomics across mutants and developmental stages reveals clusters of co-regulated genes with shared cis elements in their promoters, whose expression can be controlled positively or negatively by different ApiAP2 gene deletions. We propose that stage-specific interactions between ApiAP2 proteins on partly overlapping sets of target genes generate the complex transcriptional network that controls the Plasmodium life cycle.
