Project description:G1P3 (IFI6) was associated with poor distance metastasis free survival (DMFS) in breast cancer cases. Therefore, we tested the hypothesis that G1P3-induced mitochondrial redox deregulation confers metastatic potentials in breast cancer cells. Our results demonstrate that coordinated action of multiple pathways elicited by G1P3-induced mtROS augment actin-containing migratory structures to promote breast cancer cell migration and invasion Comparative network analysis identified upregulation of antioxidant, acin remodeling, and EMT networks in G1P3 overexpressing MCF-7 cells.

Pan-cancer analysis of IFI6 and its tumor promotor effects in esophageal squamous cell carcinoma

Project description:IFI6 is one of the ISG family, which is considered to play an important role in signal transduction pathway and immune response. However, there are few reports on IFI6, and only a few studies have reported that IFI6 may play a role in the occurrence and development of some tumors. We downloaded normal tissue and tumor tissue information from GTEx, TCGA and CCLE, and used survival analysis to study the relationship between IFI6 expression and OS, DSS, etc. We found that IFI6 was expressed in a variety of tumors, and the expression of individual tumors such as LUAD was very high. In addition, we also studied the relationship between IFI6 and immune cell infiltration enrichment, tumor immune microenvironment, methylation and so on. Furthermore, the high expression level of IFI6 was verified in esophageal squamous cell carcinoma (ESCC). In ESCC cells, IFI6 overexpression significantly promoted cell proliferation and growth, and inhibited cell apoptosis. In the lncRNA sequencing analysis, we found that IFI6 could regulate the expression of multiple genes, including HNRNPC, PEMT, INTS7, and several pathways, such as VEGF signaling pathway, and PPAR signaling pathway. Our study provides new insights into the role of IFI6 in pan-cancer. IFI6 may represent a novel target for the treatment of esophageal squamous cell carcinoma.

RNA-seq analysis of Metastatic and Localized human Pancreatic Neuroendocrine Tumors

Project description:We report the application of single-molecule-based sequencing technology for high-throughput profiling of RNAs in human pancreatic neuroendocrine tumor (PNET) tissues. We analyzed the transcriptome maps of primary tumor sites from patients that presented metastasis or not (localized). This analysis revelaed that the immune cell and pathway-related molecular signatures were enriched in metastatic compared to localized tumors. Given the wide change of gene expression in metastatic PNETs, we analyzed the RNA-seq data using a L1000 drug-induced signature to identify pharmacologic agents that might preferentially target metastatic disease. In a separate study, we analyzed our RNA-seq data to understand the molecular basis of sexual dimorphisms in PNETs. We found that PNETs are associated with the emergence of unique sex-specific transcriptomic differences that are not observed in non-neoplastic pancreatic islet tissues.

Project description:BACKGROUND:Redox deregulations are ubiquitous in cancer cells. However, the role of mitochondrial redox deregulation in metastasis remains unclear. In breast cancer, upregulation of mitochondrial antiapoptotic protein G1P3 (IFI6) was associated with poor distance metastasis-free survival (DMFS). Therefore, we tested the hypothesis that G1P3-induced mitochondrial redox deregulation confers metastatic potentials in breast cancer cells. METHODS:Cell migration and invasion assays; confocal and immunofluorescence microscopy; and Illumina HumanHT-12 BeadChip to assess gene expression. RESULTS:Consequent to its localisation on inner-mitochondrial membrane, mtROS were higher in G1P3-expressing cells (MCF-7G1P3). G1P3-overexpressing cells migrated and invaded faster than the vector controls with increased number of filopodia and F-actin bundles (p ≤ 0.05). mtROS suppression with H2O2 scavengers and mitochondrial-specific antioxidants significantly decreased migratory structures and reversed G1P3-induced migration and invasion (p ≤ 0.05). Knocking down G1P3 decreased both migration and migratory structures in MCF-7G1P3 cells. Moreover, gene networks involved in redox regulation, metastasis and actin remodelling were upregulated in MCF-7G1P3 cells. CONCLUSIONS:G1P3-induced mtROS have a direct role in migratory structure formation and nuclear gene expression to promote breast cancer cell metastasis. Therefore, interrupting mitochondrial functions of G1P3 may improve clinical outcomes in breast cancer patients.

Project description:Detection of viral infection by pattern-recognition receptors triggers production of interferon. Secreted interferon binds to cognate receptors, triggering JAK/STAT signaling, resulting in the transcription and production of hundreds of interferon-stimulated genes (ISGs). Our lab identified interferon alpha inducible protein 6 (IFI6) as an ISG that potently suppresses replication of viruses from the Flavivirus genus. To test whether the inhibitory effects of IFI6 were due to activating expression of other antiviral ISGs, we overexpressed IFI6 and a control vector and examined global transcription using RNA-Seq.

Analysis of mRNA expression in human skin fibroblasts WS1 cells between IFI6-overexpression and control upon irradiation.

Project description:This study aims to compared mRNA expression between IFI6-overexpressed WS1 cells and NC by RNA-Seq. In this study, WS1 cells were divided into two groups and each group had four repeats, one group was infected by IFI6-overexpressed lentivirus and another was infected by NC lentivirus. The two groups of cells were then irradiated with a single 5 Gy dose of radiation at a fixed rate of 1.15 Gy/min using an X-ray linear accelerator (Rad Source Technologies Inc., Suwanee, GA, USA). 24 hours post irradiation, WS1 cells from IFI6-overexpression and NC were collected and subjected to mRNA expression analysis.
