Project description:Acute myeloid leukemia (AML) represent a series of hemopathies characterized by the clonal expansion in bone marrow and blood of immature myeloid cells blocked in differentiation, called blasts. Despite significant supportive care progresses, few medical discoveries radically changed LAM prognosis (5 year survival rate of 30%). An aggressive conventional chemotherapy is used to kill AML cells. However, many patients relapse mainly due to the persistence of rare AML cells able to re-initiate the disease. Quiescence and protection against environmental stresses (chemoresistance) are major characteristics of these cells. Drug efflux leading to chemoresistance is observed by Side Population (SP) detection through ABC transporter activation. So, AML blasts presenting SP functionality could represent a chemoresistant population. This SP phenotype is essentially acquired by circulating AML blasts after contact with medullar mesenchymal stromal cells (MSC). A transcriptome analysis of AML blasts sorted according to their SP functionality or not (Non SP cells or Main Population, ie MP) after co-culture on MSCs was realized.

Side Population Analysis in Clear Cell Renal Cell Carcinoma

Project description:Cancer stem cells have an important role in tumour biology. While their identity in haematological malignancies is clearly defined, stem cell identity remains elusive in some solid tumours. Clear cell renal cell carcinoma (ccRCC) represents the most common form of kidney cancer, but the identity or existence of ccRCC stem cells remains unknown. We aimed to discern their existence using the widely utilised side population approach in ccRCC cell lines. In all cells tested, a well-defined side population was identified, and cell-based assays suggested stem-like properties. However, limiting dilution assays revealed comparable tumour initiating abilities and tumour histology of side and non-side populations, and single cell RNA-sequencing revealed minimal differences between these populations. The results indicate that the side population approach is not sufficient for cancer stem cell discovery in ccRCC.

Blast wave effect on brain proteome (rat hippocampus)

Project description:These data show differences in up- and down-regulation for protein abundances in the hippocampus of double blast vs. sham rats. Tandem mass tags (TMT)-MS results showed 136 up-regulated and 94 down-regulated proteins between the two groups. These TMT-MS findings revealed changes never described before in blast studies. In the absence of behavioral changes, these proteomic data further support the existence of an asymptomatic blast-induced molecular altered status (ABIMAS) associated with specific protein changes in the rat hippocampus.

Single-cell RNA sequencing reveals the induction of novel myeloid and myeloid-associated cell populations in visceral fat with long-term obesity

Project description:Macrophages and other immune cells are important contributors to obesity-associated inflammation; however, the cellular identities of these specific populations remain unknown. In this study, we identified individual populations of myeloid cells found in mouse epididymal/visceral adipose tissue by single-cell RNA sequencing, immunofluorescence, and flow cytometry. Multiple canonical correlation analysis identified 11 unique myeloid and myeloid-associate cell populations. In obese mice, we detected an increased percentage of monocyte-derived pro-inflammatory cells expressing CD9 and TREM-2, as well as significantly decreased percentages of multiple cell populations, including tissue-resident cells expressing LYVE1, Mafb, and CD206. We have identified and validated a novel myeloid/macrophage population defined by Ly6a expression, exhibiting both myeloid and mesenchymal characteristics, with both hematopoietic and pro-fibrotic markers. Our mouse adipose tissue myeloid cell atlas provides an important resource to target obesity-associated inflammation and fibrosis.
