Project description:In this study, we found that the highly expressed miRNAs appear in EPDELNs (Edible plant-derived exosome-like nanoparticles) have the potential to regulate human mRNA in a cross-kingdom manner. Target prediction and functional analysis revealed that highly expressed miRNAs were closely associated with inflammatory responses and cancer-related pathways. Therefore, we hypothesized that miRNAs in EPDELNs may be a crucial factor behind the regulatory function of EPDELNs.

Project description:MicroRNAs (miRNAs) are small non-coding RNAs between 18-23nts in size which regulate the translation and stability of target mRNAs. miRNAs are present in dietary plants and are conventionally thought to be degraded during the gastrointestinal digestion process. Recent reports suggest that a few dietary microRNAs may exhibit resistance to this process, enter systemic circulation and exert biological effects on animal physiology, currently known as cross-kingdom regulation. However, such horizontal transfer of miRNAs via different kingdoms is highly likely for miRNAs that are present intrinsic extracellular vesicles which increases their bioavailability. These vesicular structures from plants are known as Exosome-like nanovesicles (ENV). ENVs have been isolated from several edible plants. ENV-derived miRNAs are probably more bioavailable and are spontaneously absorbed in intestinal epithelium to suppress target transcripts in human/microbial/viral kingdoms. Such cross-kingdom regulation exhibited by ENV-miRNAs, if properly investigated and validated, may aid in the development of non-toxic and cost-effective therapeutics to treat human diseases. In this line, we purified ENVs from four edible plants (Soy bean, ginger, amla and turmeric). Small RNA population from these ENVs were isolated and profiled through small RNA sequencing to identify ENV-associated miRNAs enriched in each species.

Project description:Edible plant-derived exosome-like nanoparticles (EPDELNs) are novel naturally occurring plant ultrastructures that are structurally similar to exosomes. Many EPDELNs have anti-inflammatory properties. MicroRNAs (miRNAs) play a critical role in mediating physiological and pathological processes in animals and plants. Although miRNAs can be selectively encapsulated in extracellular vesicles, little is known about their expression and function in EPDELNs. In this study, we isolated nanovesicles from 11 edible fruits and vegetables and subjected the corresponding EPDELN small RNA libraries to Illumina sequencing. We identified a total of 418 miRNAs-32 to 127 per species-from the 11 EPDELN samples. Target prediction and functional analyses revealed that highly expressed miRNAs were closely associated with the inflammatory response and cancer-related pathways. The 418 miRNAs could be divided into three classes according to their EPDELN distributions: 26 "frequent" miRNAs (FMs), 39 "moderately present" miRNAs (MPMs), and 353 "rare" miRNAs (RMs). FMs were represented by fewer miRNA species than RMs but had a significantly higher cumulative expression level. Taken together, our in vitro results indicate that miRNAs in EPDELNs have the potential to regulate human mRNA.

Project description:Introduction: microRNAs are promising candidate breast cancer biomarkers due to their cancer-specific expression profiles. However, efforts to develop circulating breast cancer biomarkers are challenged by the heterogeneity of microRNAs in the blood. To overcome this challenge, we aimed to develop a molecular profile of microRNAs specifically secreted from breast cancer cells. Our first step towards this direction relates to capturing and analyzing the contents of exosomes, which are small secretory vesicles that selectively encapsulate microRNAs indicative of their cell of origin. To our knowledge, circulating exosome microRNAs have not been well evaluated as biomarkers for breast cancer diagnosis or monitoring. Methods: Exosomes were collected from the conditioned media of human breast cancer cell lines, mouse plasma of patient-derived orthotopic xenograft models (PDX), and human plasma samples. Exosomes were verified by electron microscopy, nanoparticle tracking analysis, and western blot. Cellular and exosome microRNAs from breast cancer cell lines were profiled by next-generation small RNA sequencing. Plasma exosome microRNA expression was analyzed by qRT-PCR analysis. Results: Small RNA sequencing and qRT-PCR analysis showed that several microRNAs are selectively encapsulated or highly enriched in breast cancer exosomes. Importantly, the selectively enriched exosome microRNA, human miR-1246, was detected at significantly higher levels in exosomes isolated from PDX mouse plasma, indicating that tumor exosome microRNAs are released into the circulation and can serve as plasma biomarkers for breast cancer. This observation was extended to human plasma samples where miR-1246 and miR-21 were detected at significantly higher levels in the plasma exosomes of 16 breast cancer patients as compared to the plasma exosomes of healthy control subjects. Receiver Operating Characteristic (ROC) curve analysis indicated that the combination of plasma exosome miR-1246 and miR-21 levels is a better indicator of breast cancer than their individual levels. Conclusions: Our results demonstrate that certain microRNA species, such as miR-21 and miR-1246, are selectively enriched in human breast cancer exosomes and significantly elevated in the plasma of breast cancer patients. These findings indicate a potential new strategy to selectively analyze plasma breast cancer microRNAs indicative of the presence of breast cancer.

Project description:Plant-based diets could be a key source of microRNAs in animals. Plant microRNAs are cross-kingdom gene expression regulators that could modulate mammalian gene expression, influencing their physiology. Therefore, it is important to identify the microRNA expression profile of plant foods in order to identify potential target genes and biological functions in the mammalian host. Next-generation sequencing was applied to identify microRNAs in RNA samples derived from nuts (walnut and almond), vegetables (spinach) and fruits (orange, apple, olive, pear, and tomato). Our data revealed that edible plant contain a large number and diverse type of microRNAs.

Project description:The increasing commercial production of engineered nanoparticles (ENPs) has led to concerns over the potential adverse impacts of these ENPs on biota in natural environments. Silver nanoparticles (AgNPs) are one of the most widely used ENPs and are expected to enter natural ecosystems. Here we examined the effects of AgNPs on germination and growth of eleven species of common wetland plants. We examined plant responses to AgNP exposure in simple pure culture experiments (direct exposure) and for seeds planted in homogenized field soils in a greenhouse experiment (soil exposure). We compared the effects of two AgNPs-20-nm polyvinylpyrrolidine-coated silver nanoparticles (PVP-AgNPs) and 6-nm gum arabic coated silver nanoparticles (GA-AgNPs)-to the effects of AgNO(3) exposure added at equivalent Ag concentrations (1, 10 or 40 mg Ag L(-1)). In the direct exposure experiments, PVP-AgNP had no effect on germination while 40 mg Ag L(-1) GA-AgNP exposure significantly reduced the germination rate of three species and enhanced the germination rate of one species. In contrast, 40 mg Ag L(-1) AgNO(3) enhanced the germination rate of five species. In general root growth was much more affected by Ag exposure than was leaf growth. The magnitude of inhibition was always greater for GA-AgNPs than for AgNO(3) and PVP-AgNPs. In the soil exposure experiment, germination effects were less pronounced. The plant growth response differed by taxa with Lolium multiflorum growing more rapidly under both AgNO(3) and GA-AgNP exposures and all other taxa having significantly reduced growth under GA-AgNP exposure. AgNO(3) did not reduce the growth of any species while PVP-AgNPs significantly inhibited the growth of only one species. Our findings suggest important new avenues of research for understanding the fate and transport of NPs in natural media, the interactions between NPs and plants, and indirect and direct effects of NPs in mixed plant communities.
