Project description:The environmental fate of the herbicide fenoxaprop-ethyl (FE) in water, sediment and water-sediment microcosm was studied and degradation products fenoxaprop (FA), ethyl-2-(4-hydroxyphenoxy)propanoate (EHPP), 2-(4-hydroxyphenoxy)propanoic acid (HPPA) and 6-chloro-2,3-dihydrobenzoxazol-2-one (CDHB) were monitored. FE, FA, EHPP and HPPA were chiral and the environmental behavior was investigated on an enantiomeric level. In water, sediment and water-sediment microcosms, fenoxaprop-ethyl degraded very fast with half-lives less than 1 day and it was found the herbicidally inactive S-enantiomer degraded faster. Fenoxaprop was the main primary degradation product which was quickly formed and the further degradation was relatively slow with half-lives of 6.4-12.4 days, and the S-enantiomer degraded faster too. EHPP, HPPA and CDHB could be found and S-EHPP and S-HPPA were degraded preferentially. The effects of microorganism and water content were investigated and it was found that the enantioselectivity was attributed to microorganisms. In sediment, the main degradation pathway of fenoxaprop-ethyl was hydrolysis and the degradation rate of fenoxaprop-ethyl increased with water content. The degradation products and enantioselectivity should be considered for the impact of fenoxaprop-ethyl on the aquatic system.

Project description:The use of aquatic environmental DNA (eDNA) to detect the presence of species depends on the seasonal activity of the species in the sampled habitat. eDNA may persist in sediments for longer than it does in water, and analysing sediment could potentially extend the seasonal window for species assessment. Using the great crested newt as a model, we compare how detection probability changes across the seasons in eDNA samples collected from both pond water and pond sediments. Detection of both aquatic and sedimentary eDNA varied through the year, peaking in the summer (July), with its lowest point in the winter (January): in all seasons, detection probability of eDNA from water exceeded that from sediment. Detection probability of eDNA also varied between study areas, and according to great crested newt habitat suitability and sediment type. As aquatic and sedimentary eDNA show the same seasonal fluctuations, the patterns observed in both sample types likely reflect current or recent presence of the target species. However, given the low detection probabilities found in the autumn and winter we would not recommend using either aquatic or sedimentary eDNA for year-round sampling without further refinement and testing of the methods.

Project description:Environmental DNA (eDNA) surveys are increasingly being used for biodiversity monitoring, principally because they are sensitive and can provide high resolution community composition data. Despite considerable progress in recent years, eDNA studies examining how different environmental sample types can affect species detectability remain rare. Comparisons of environmental samples are especially important for providing best practice guidance on early detection and subsequent mitigation of non-indigenous species. Here we used eDNA metabarcoding of COI (cytochrome c oxidase subunit I) and 18S (nuclear small subunit ribosomal DNA) genes to compare community composition between sediment and water samples in artificial coastal sites across the United Kingdom. We first detected markedly different communities and a consistently greater number of distinct operational taxonomic units in sediment compared to water. We then compared our eDNA datasets with previously published rapid assessment biodiversity surveys and found excellent concordance among the different survey techniques. Finally, our eDNA surveys detected many non-indigenous species, including several newly introduced species, highlighting the utility of eDNA metabarcoding for both early detection and temporal / spatial monitoring of non-indigenous species. We conclude that careful consideration on environmental sample type is needed when conducting eDNA surveys, especially for studies assessing community change.

Project description:Despite representing one of the largest biomes on earth, biodiversity of the deep seafloor is still poorly known. Environmental DNA metabarcoding offers prospects for fast inventories and surveys, yet requires standardized sampling approaches and careful choice of environmental substrate. Here, we aimed to optimize the genetic assessment of prokaryote (16S), protistan (18S V4), and metazoan (18S V1-V2, COI) communities, by evaluating sampling strategies for sediment and aboveground water, deployed simultaneously at one deep-sea site. For sediment, while size-class sorting through sieving had no significant effect on total detected alpha diversity and resolved similar taxonomic compositions at the phylum level for all markers studied, it effectively increased the detection of meiofauna phyla. For water, large volumes obtained from an in situ pump (~ 6000 L) detected significantly more metazoan diversity than 7.5 L collected in sampling boxes. However, the pump being limited by larger mesh sizes (> 20 µm), only captured a fraction of microbial diversity, while sampling boxes allowed access to the pico- and nanoplankton. More importantly, communities characterized by aboveground water samples significantly differed from those characterized by sediment, whatever volume used, and both sample types only shared between 3 and 8% of molecular units. Together, these results underline that sediment sieving may be recommended when targeting metazoans, and aboveground water does not represent an alternative to sediment sampling for inventories of benthic diversity.

Project description:More than 2300 sediment pore water distribution coefficients (KPCBi ds) of 93 polychlorinated biphenyls (PCBs) were measured and modeled from sediments from Indiana Harbor and Ship Canal. KPCBi ds were calculated from previously reported bulk sediment values and newly analyzed pore water. PCBs in pore waters were measured using SPME PDMS-fiber and ?PCB ranged from 41 to 1500 ng L(-1). The resulting KPCBi ds were ~1 log unit lower in comparison to other reported values. A simple model for the KPCBi d consisted of the product of the organic carbon fraction and the octanol-water partition coefficient and provided an excellent prediction for the measured values, with a mean square error of 0.09 ± 0.06. Although black carbon content is very high in these sediments and was expected to play an important role in the distribution of PCBs, no improvement was obtained when a two-carbon model was used.

Project description:Deep-water renewal (DWR) events are characterized in the Strait of Georgia, Canada using 11 years of real-time physical and chemical oceanographic data and seafloor videos. At least 6 DWRs occur per year at 300 m water depth and each event continues for over 3 days. They initiate during neap tides and are associated with increased turbidity. In the spring, DWRs introduce cold, oxygenated and nutrient-poor waters, and in the fall they introduce warm, oxygen-depleted, nutrient-rich and saline waters. Although the timing and magnitude of DWRs differ from year to year, we demonstrate that they are not restricted to two seasons, but continue throughout the year. High-resolution videos of DWRs show that these events comprise a plume of high suspended sediment concentration that flows parallel to the basin axis and deposits approximately 1.5 cm per event.
