Project description:The present study aimed to compare the miRNA expression profile in individuals with chronic (CP) and aggressive (AP) periodontitis. Eighteen non-smokers individuals (CP = 9 and AP = 9) without any history of systemic diseases or previous periodontal therapy were selected at the Clinics of Periodontology from the Federal University of Minas Gerais. Gingival tissue samples were collected during the initial periodontal therapy. miRNAs were isolated and expression patterns of 754 miRNAs were assessed with a quantitative miRNA PCR array. miRNAs expression profiles were compared between CP and AP groups. No differences were observed in the miRNAs expression profiles between CP and AP groups (p>0.05). According to miRNA microarray analyses, the most expressed miRNAs in both groups were hsa-miR-1274b, hsa-let-7b, hsa-miR-24, hsa-miR-19b, hsa-miR-720, hsa-miR-126, hsa-miR-17 and hsa-miR-21. Findings suggested that there were no differences in miRNAs expression profiles between chronic and aggressive forms of periodontitis. The overexperssion of especific miRNAs could provide insights on the pathogenesis of both forms of the disease.

Project description:Periodontitis is a chronic inflammatory disease resulting from a dysbiosis of the dental biofilm and a dysregulated host response in susceptible individuals. It is characterized by periodontal attachment destruction, bone resorption, and eventual tooth loss. Salivary biomarkers have been sought to predict and prevent periodontitis. This comparative study analyzed the salivary proteome of 30 individuals with chronic periodontitis (CP) and 10 with periodontal health (PH), and correlated specific proteins with clinical parameters of disease by using mass spectrometry. Stimulated whole saliva was obtained and pooled for 5 healthy controls and 15 CP patients, precipitated with TCA, digested enzymatically with trypsin and analyzed by an LTQ Orbitrap Velos equipped with a nanoelectrospray ion source. A wide range of salivary proteins of various functions was significantly reduced in CP individuals, whereas salivary acidic proline-rich phosphoprotein, submaxillary gland androgen-regulated protein, histatin, fatty acid-binding protein, thioredoxin, and cystatin were predominant in diseased patients and correlated significantly with signs of periodontal attachment loss and inflammation. Specific salivary proteins were associated with PH and CP. These differences in salivary proteome profiles may contribute to the identification of disease indicators or signatures and the improvement of periodontal diagnosis.

Project description:Aggressive periodontitis (AP; Grade C, Stage 3-4) is rapid destructive condition of periodontium triggered by an imbalanced immune reaction with dysbiosis of periodontal pathogens. AP is characterized by extensive tooth loss in early ages due to its severe nature and rapid progression, significantly impacting the affected individual's quality of life. Despite the severity and refractoriness of the disease, the precise pathophysiology of this hyper-reactive phenotype remains unclear. In this study, we dissected the distinct immunological features in AP by establishing the comprehensive human gingival cell atlas on individuals with AP, chronic periodontitis (CP), and healthy controls (HC). Iterative clustering analysis based on the single-cell transcriptome revealed the distinct populations in B and plasma cells upon the periodontal conditions. AP group exhibited a significant rise in B cells including a unique cell cluster dominantly expressing ID3, a key suppressor of plasma cell development. This observation is coordinated with the impaired phenotypes of antibody class switching in AP patients. Remarkably, the secreted IgM gingival crevicular fluid from AP patients exhibited the autoreactivity to gingival tissues, suggesting a potential autoimmune mechanism that involved in AP pathogenesis. These findings highlight distinct immune cell and antibody profiles in AP, providing the insights into the groundwork for personalized monitoring and proactive intervention approaches to mitigate irreversible tissue damage.

Project description:Gene expressions relate to the pathogenesis of periodontitis and have a crucial role in local tissue destruction and susceptibility to the disease. The aims of the present study were to explore comprehensive gene expressions/transcriptomes in periodontitis-affected gingival tissues, and to identify specific biological processes. The purpose of the present study was 1) to compare comprehensive gene expression/transcriptomes of periodontitis-affected gingival tissues with those of healthy tissues by using microarray and data mining technologies, and 2) to analyze significantly differentially expressed genes which belong to pathological pathways in periodontitis by qRT-PCR. Two distinct gingival samples including healthy and periodontal-affected gingiva were taken from 3 patients with severe chronic periodontitis. Total RNAs from 6 gingival tissue samples were used for microarray and data-mining analyses. Comparisons, gene ontology, and pathway frequency analyses were performed and identified significant biological pathways in periodontitis. Quantitative reverse transcription real-time polymerase chain reaction (qRT-PCR) analyse using 14 chronic periodontitis patients including 3 patients listed above and 14 healthy individuals showed 9 differentially expressed genes in leukocyte migration and cell communication pathways.

Project description:We report the application of high throughput Illumina sequencing for profiling of small RNAs in saliva of patients who were diagnosed with chronic periodontitis as compared to healthy controls. To date, there is no published literature on salivary microRNA profiling done using the high throughput next-generation sequencing analysis in patients diagnosed with chronic periodontitis. Also, this is the first study of its kind done in an Indian population. The objectives of the study were to profile microRNAs expressed in saliva of patients diagnosed with chronic periodontitis, to identify differentially expressed microRNAs between chronic periodontitis and healthy patients and to identify putative salivary microRNAs which can serve as biomarkers for periodontal disease.
