Project description:The aim of this study was to determine how nitrogen repletion affected the genomic cell response of a Saccharomyces cerevisiae wine yeast strain, in particular within the first hour following relief from nutrient starvation. We found almost 4000 genes induced or repressed sometimes within minutes of nutrient changes. Some of the transcriptional responses to nitrogen depended on the TOR pathway which control positively ribosomal protein genes, amino acid and purine biosynthesis or amino acid permease genes and negatively stress-response genes, RTG specific TCA cycle genes and NCR sensitive genes. Some unexpected transcriptional responses concerned all the glycolytic genes, the starch and glucose metabolism and citrate cycle related genes which were down-regulated, as well as genes from the lipid metabolism.

Specific features of the transcriptomic response to nitrogen starvation in methylotrophic yeast Komagataella phaffi

Project description:Non-conventional methylotrophic yeast Komagataella phaffii is an important production host in biotechnology and an emerging model organism. In this work, we studied K. phaffii response to nitrogen starvation during cultivation in media with methanol as the sole carbon source. The results were compared with well-established model yeast Saccharomyces cerevisiae. Some of the observed effects of nitrogen starvation in K. phaffii were similar to those in S. cerevisiae, despite this yeast does not have metabolic pathway for methanol utilization. The effects include activation of autophagy, transport and catabolism of nitrogen-containing compounds, interconversions of amino acids, and biosynthesis of fatty acids. K. phaffii cells also demonstrated specific response to nitrogen starvation including suppression of genes involved in methanol metabolism and other peroxisomal processes and activation of purine catabolism genes.

Project description:The protein kinase Ime2 is known to have an important role in meiosis in the yeast Saccharomyces cerevisiae. However, the Neurospora crassa IME2 homolog functions in self/nonself recognition as well as in the development of fungal sexual structures called protoperithecia. In N. crassa, protoperithecia are induced upon nitrogen starvation. We were interested in gene expression differences between an ime-2 deletion strain and a wild type strain, and due to the role of ime-2 during sexual development, carried out these arrays on media that was lacking in nitrogen.

Project description:In conditions of nitrogen limitation, Saccharomyces cerevisiae strains differ in their fermentation capacities, due to differences in their nitrogen requirements. A population of 133 individuals from the F2 segregant population from a cross between two strains with different nitrogen requirements for efficient fermentation has been analyzed for their fermentation capacities. Two groups of 15 strains were defined, one group of High and one of Low Nitrogen requirement. These two groups are compared in order to detect genomic regions involved in the differences of nitrogen requirement. We used a custom isothermal array that has been designed for the detection of SNP at 6317 position on RM11.1a genome sequence http://www.broadinstitute.org/annotation/genome/saccharomyces_cerevisiae.3/Home.html) and obtained from the comparison with the genome sequence of strain Saccharomyces P3-D5.
