Project description:Purpose: The goal of this study was to assess the differential gene expression between control and leptin treated pituitary rostral pars distalis' of the tilapia (Oreochromis mossambicus). Methods: Pituitary mRNA profiles from control (PBS) and leptin (100nM) treated tilapia (Oreochromis mossambicus) pituitary rostral pars disatalis were generated by deep sequencing using Illumina MiSeq. The sequence reads that passed quality filters were analyzed at the transcript abundance level using ANOVA (ANOVA) and BowTie2 followed by Cufflinks. Results: Differential gene expression identified 1995 genes that were differentially expressed in the pituitary rostral pars distalis treated with leptin compared to PBS treated controls. These genes were indicated to be functionally involved in numerous biological processes including glycolyiss. Conclusions: Our study represents the first detailed pituitary rostral pars distalis transcriptomes in the tilapia and identifies leptin as a major stimulator of glycolysis at the cellular level in teleost fishes.

Project description:We assessed if leptin, a cytokine hormone known to enhance energy expenditure by promoting lipid and carbohydrate catabolism in response to physiologic stress, might directly regulate cellular glycolysis. A transcriptomic analysis of prolactin cells in the tilapia (Oreochromis mossambicus) pituitary rostral pars distalis (RPD) revealed that recombinant leptin (rtLep) differentially regulates 1,995 genes, in vitro. Machine learning algorithms and clustering analyses show leptin influences numerous cellular gene networks including metabolism; protein processing, transport, and metabolism; cell cycle and the hypoxia response. Leptin stimulates transcript abundance of the glycolytic enzyme glyceraldehyde-3-phosphate dehydrogenase (gapdh) in a covariate manner to the hypoxic stress gene network. Orthogonal tests confirm that rtLepA dose-dependently increases gapdh gene expression in the RPD along with transcript abundance of 6-phosphofructo-1-kinase (pfk1), the rate limiting glycolytic enzyme. Functional testing demonstrated that leptin stimulates PFK activity and glycolytic output, while Stattic (a STAT3 blocker) was sufficient to suppress these responses, indicating leptin stimulates glycolysis through a STAT3-dependent mechanism. Leptin also stimulated pfk1 gene expression and lactate production in primary hepatocyte incubations in a similar manner to those shown for the pituitary RPD. This work characterizes a critical metabolic action of leptin to directly stimulate glycolysis across tissue types in a teleost model system, and suggest that leptin may promote energy expenditure, in part, by stimulating glycolysis. These data in a teleost fish, suggest that one of leptin's ancient, highly-conserved functions among vertebrates may be stimulation of glycolysis to facilitate the energetic needs associated with various stressors.

Project description:Leptin, a hormone produced primarily by adipose tissue, plays a role in both energy homeostasis and reproduction, and is required in early pregnancy. Leptin stimulates metalloproteinase activity in cultured human trophoblast and stimulates invasiveness of cultured mouse trophoblast. The goal of the present study was to examine molecular mechanisms of this function in primary cultures of mouse trophoblast. Leptin was found to stimulate the phosphorylation of MEK, but not STAT3.. It also increased levels of the protein SOCS3. The ability of leptin to stimulate metalloproteinase activity was blocked by the MEK inhibitor PD98059, but also by the vehicle inhibitor DMSO. Microarray analysis revealed that leptin stimulated some genes associated with cell motility, such as Stmn1. In addition, leptin appeared to inhibit changes in gene expression associated with terminal differentiation of trophoblast giant cells, including inhibition of members of the TGFÃ signaling pathway and of genes associated with endoreduplication. However, feulgen staining revealed a loss of cells with low ploidy. We conclude that leptin may be promoting trophoblast invasion by maintaining cells in an intermediate stage of differentiation. There were two treatments: control (serum-free medium) and recombinant mouse leptin (50 ng/mL). RNA was collected at two time points, 1 hour and 24 hours, for the controls. RNA was collected at three time points after leptin treatment: 1 hour, 6 hours, and 24 hours. Thus, there were 5 samples for each experiment. The experiment was repeated four times, for a total of 20 arrays.

Project description:Undernutrition increases susceptibility to diarrheal diseases. The adipocytokine leptin imparts protection from amebiasis. We tested the role of leptin signaling in cellular resistance to E. histolytica cytotoxicity in HEK cells transfected to express the leptin receptor. Protection from amebic killing was conferred by the leptin receptor. It required activation of the transcriptional regulator STAT3 by the leptin receptor, as mutation of the STAT3 activation domain of the receptor, or addition of a STAT3 small-molecule inhibitor, reversed protection. In contrast, a leptin receptor containing a common polymorphism (Q223R) known to increase susceptibility to amebiasis in humans provided significantly less protection. Consistent with the importance of STAT3, the Q223R polymorphism decreased l leptin-dependent STAT3 activation by 21% relative to the WT receptor (P=0.035). Microarray analysis identified potential downstream effectors of STAT3-mediated protection, most importantly TRIB1 and SOCS3, which appear to having opposing roles in the regulation of E. histolytica induced apoptosis. Together these data demonstrated that leptin increased the resistance of host cells to E. histolytica cytotoxicity via a STAT3-dependent mechanism. Additionally we found that the Q223R polymorphism in the leptin receptor, known to increase susceptibility to E. histolytica infection, decreased STAT3 activation and decreased host resistance to amebic cytotoxicity. This is the first demonstration of a host-signaling pathway that restricts amebic pathogenesis. Additionally, this finding represents an important advance in our mechanistic understanding of the role of leptin in the relationship between undernutrition and increased susceptibility to infection.

Project description:Signal Transducer and Activator of Transcription 3 (STAT3) is considered as an oncogene being constitutively activated in a wide variety of primary human tumours, which often become addicted to its activity. Using primary fibroblasts derived from a knock-in mouse expressing only the constitutively active form STAT3-C as well as STAT3-dependent tumor cell lines MDA-MB468, DU145 and SKBR3, we provide evidence that STAT3 can act as a central regulator of cell metabolism. STAT3C/C MEFs are resistant to apoptosis and senescence and they show reduced mitochondrial activity but activate aerobic glycolysis, as shown by enhanced expression of master regulators of glycolysis such as PDK-1 and HIF-1α, and by increased lactate production, glucose avidity and sensitivity to glycolysis inhibitors. HIF-1α is up-regulated in the STAT3C/C MEFs and is responsible for the glycolytic phenotype. Moreover, inhibition of STAT3 activity in STAT3-addicted tumour cell lines restores mitochondrial activity and down-regulates glycolytic metabolism, preceding the induction of massive apoptosis. Thus, the essential role observed for STAT3 in so many different cancer cell types may be partly explained by its ability to act as a molecular switch of cellular metabolism triggering abnormal cell survival/proliferation. Three biological replicates of MEFs from mice with constitutively active Stat3 are compared to three biological replicates of wild-type MEFs.

Project description:Liver fibrosis occurs during chronic liver disease. Advanced liver fibrosis results in cirrhosis, liver failure and often requires liver transplantation. However, due to the lack of human models, mechanisms underlining the pathogenesis of liver fibrosis remain unclear. Recent studies implicated a central role of deranged lipid metabolism in its pathogenesis. In this study, we generated LEPTIN-deficient (LEPTIN-/-) pigs using zinc finger nuclease technology to investigate the mechanisms of liver fibrosis associated with obesity. The LEPTIN-/- pigs showed increased body fat and significant insulin resistance by 12 months of age. To resemble non-alcoholic fatty liver disease (NAFLD) patients, LEPTIN-/- pig developed the phenotypic features of fatty liver, non-alcoholic steatohepatitis (NASH) and hepatic fibrosis with age. Meanwhile, LEPTIN absence reduced phosphorylation of JAK2-STAT3 and AMPK. The alteration of JAK2-STAT3 enhanced fatty acid β-oxidation, whereas inactivation of AMPK led to mitochondrial autophagy, and both contributed to increased oxidative stress in hepatocytes. Although Leptin deletion in the rat liver altered JAK2-STAT3 phosphorylation, it activated the AMPK pathway and prevented liver fibrogenesis in contrast with the LEPTIN-/- pig. To our knowledge, the LEPTIN-/- pig provides the first model recapitulating the full pathogenesis of NAFLD and its progression toward liver fibrosis. The activity of AMPK signaling pathway suggests a potential target for development of new strategies for the diagnosis and treatment of NAFLD.
