Mass spectrometry analysis of synaptic proteome of adult Fmr1 KO and wild-type mice.

Project description:Fragile X syndrome (FXS) is the most common monogenetic cause of inherited intellectual disability and autism in humans. One of the well-characterized molecular phenotypes of Fmr1 KO mice, a model of FXS, is increased translation of synaptic proteins. Although this upregulation stabilizes in the adulthood, abnormalities during the critical period of plasticity have long-term effects on circuit formation and synaptic properties. Using high-resolution quantitative proteomics of synaptoneurosomes isolated from the adult, developed brains of Fmr1 KO mice, we show differential abundance of proteins regulating postsynaptic receptor activity of glutamatergic synapse. This work includes proteomic dataset that was acquired and analyzed to quantify changes in the abundance of proteins in synaptoneurosomes (basal state, unstimulated and in vitro NMDA-R stimulated) isolated from Fmr1 KO mice and their wild-type littermates.

Differential gene expression in the airway epithelium of beta6 ko mice chronically challenged with OVA

Project description:Expression microarrays identified 119 genes that were significantly differentially expressed in the epithelium of WT and M-NM-26 KO mice after saline or chronic allergen challenge. PAM clustering revealed two interesting clusters (6 and 8) that we had not identified in previous comparisons by whole lung microarrays. Cluster 6 genes were low at baseline in both WT and M-NM-26 KO mice and were increased in WT but not in M-NM-26 KO mice after chronic allergen challenge such as Mcpt1. Cluster 8 genes were increased at baseline in M-NM-26 KO mice, and included 6 mast cell related genes (cma1, mcpt4, cpa3, mcpt6, tpsab1, il1rl1). The most informative differentially expressed genes identified in microarrays of the epithelial microenvironment were not epithelial genes, but mast cell genes. Control and beta6 ko mice (wt-S (4), wt-OVA (4), ko-S (4), ko-OVA (4)) were sensitized and challenged with OVA or saline, airway epithelium were brush-harvested.

Maternally inherited piRNAs silence transposons during Drosophila embryogenesis

Project description:The piRNA pathway controls transposon expression in animal germ cells, thereby ensuring genome stability over generations. piRNAs are maternally deposited and required for proper transposon silencing in adult offspring. However, a long-standing question in the field is the precise function of maternally deposited piRNAs and its associated factors during embryogenesis. Here, we probe the spatio-temporal expression patterns of several piRNA pathway components during early stages of development. Amongst those, factors required for transcriptional gene silencing (TGS) showed ubiquitous abundance in somatic and pole cells throughout the first half of embryogenesis. We further analysed the transcriptomes of various embryo stages and correlated these with the presence of selected chromatin marks. We found that a number of transposon families show bursts of transcription during early embryonic stages. Transposons heavily targeted by maternally deposited piRNAs accumulated repressive chromatin marks following their spike in expression. Furthermore, depletion of maternally deposited Piwi protein in early embryos resulted in increased expression of transposons targeted by inherited piRNAs and was accompanied by a strong loss of repressive chromatin marks at coding sequences. Overall, our data suggests a pivotal role for the piRNA pathway in transposon defence during Drosophila embryogenesis in somatic cells.
