Project description:We have previously reported that the responses of tobacco tissues treated with AaGlucan are similar to those for laminarin [1]. To better understand the overall responses and signaling pathways in tobacco, we have identified here AaGlucan and laminarin early response genes, using a custom 16K BY-2 microarray [2,3]. BY-2 cells were treated with laminarin or an AaGlucan-enriched fraction prepared from A. alternata 102 culture-medium and subjected to microarray analysis. Project description:We compared the gene expression stimulated with fungal extracts from Aspergillus (A.) fumigatus, Alternaria (A.) alternata, or Penicillium (P.) notatum in NCI-H292 (a human bronchial epithelial cell line) to search Allergic bronchopulmonary mycosis (ABPM)-related genes. We identified a mucin-related MUC5AC gene, the expression of which was selectively induced by A. fumigatus. Total RNA from NCI-H292 cells stimulated for 24 h with the A. fumigatus, A. alternata, or P. notatum fungi extracts was extracted and subjected to microarray analysis. Each experiments were perfomed once for each stimulus.

Project description:This present study is the first to investigate the global changes in host gene expression during the interaction of human bronchial epithelial cells and live Alternaria spores. Human bronchial epithelial cells (BEAS2-B) were exposed to spores or media alone for 24 hours. RNA was collected from three biological replicates/treatment and used to assess changes in gene expression patterns using Affymetrix Human Genome U133 Plus 2.0 Arrays. Interestingly, many cytokine/chemokine immune response genes were upregulated. Genes involved in cell death, retinoic acid signaling, TLR3, and interferon response pathways were also significantly upregulated. Project description:Trichoderma atroviride is a fungus capable of establishing symbiotic relationships with plants, however, its main lifestyle is a saprophyte. Due to these characteristics, it must face a great quantity of microorganisms, and be able to compete for nutrients. T. atroviride is considered a necrotrophic mycoparasite and has developed the ability to kill other organisms and obtain nutrients from them. The object of this work is to explore the role of small RNAs in mycoparasitism. To this end, we obtained small RNA-Seq libraries from the interactions of T. atroviride against Alternaria alternata. The libraries were obtained from three stages during mycoparasitsm: before contact (BC), contact (C), and after contact (AC).
