Project description:The study investigated differences in gene expression profiles of peripheral blood nuclear cells sampled from healthy and infected with SRLV goats, belonging to two breeds. Due to the nature of the lentiviral infection we hypothesized that the virus could induce genes expression regulation. We put forward the hypothesis that if gene expression differences exist, they should be revealed in studies conducted on custom-made Capra hircus Gene Expression Array. The results revealed that SRLV infection results in regulation of several genes in goat’s peripheral blood nuclear cells.

Project description:The immune response to a viral antigen causes inflammatory cell infiltration to the tissue, which creates a suitable environment for the replication of the virus in macrophages, and the recruitment of more monocytes to the site of infection, or latently infected monocytes. The aim of the study was to analyze the transcriptomic profile of peripheral blood nuclear cells isolated from SRLV-seropositive and SRLV-negative goats at the peak of their first lactation. SRLV-seropositive goats were probably infected via colostrum. Custom transcriptomic microarrays for goats were designed and developed, namely the Capra hircus gene expression array, which features ~50,000 unique transcripts per microarray. Only four genes were differentially expressed, with up-regulated expression of the GIMAP2, SSC5D and SETX genes, and down-regulated expression of the GPR37 gene in SRLV-seropositive vs. SRLV-seronegative goats. However, in an RT-qPCR analysis, the result for the SETX gene was not confirmed. The differences in the expressions of the studied genes indicate an active inflammatory process in the SRLV-seropositive goats at the early stage of infection.

Project description:In this study, m6A-modified Methylated RNA immunoprecipitation sequencing (MeRIP-seq) and transcriptome sequencing (RNA-seq) were used to identify the key genes with m6A modification during mammary gland development and lactation in dairy goats. The results showed that m6A methylation occurred at 3,927 loci, which were significantly enriched in the 3' untranslated coding region(3’UTR) and the termination codon region. In the early stage (E) and peak stage (P) of lactation, m6A methylation occurred extensively in mammary tissues, and a total of 725 differentially expressed m6A-modified genes were obtained, all negatively correlated with mRNA expression. In addition, gene ontology (GO) enrichment and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis showed that different methylated genes were mainly involved in the growth and apoptosis of mammary epithelial cells through signaling pathways related to mammary gland development and lactation, such as the mitogen-activated protein kinase (MAPK) and phospholipase D pathways.

Project description:Purpose: RNA-sequencing (RNA-seq) was used to identify the changes in gene expression in goats carrying high (HPL) and low (LPL) proviral load of SRLV. For the first time, differentially expressed transcriptome profiles in these two group of animals were identified, and information about genes and mechanisms, and relevant pathways inferred via bioinformatics analyses. The results provided unique insights for further exploring and understanding the host responses to SRLV infection in goats. Results: Data enabled to identify 1434 significant differentially expressed genes between HPL and LPL goats. Of these genes, 571 were upregulated and the remaining 863 genes were downregulated in HPL goats. GO enrichment analysis showed that the identified DEGs were found to have significantly enriched regulation of signaling receptor activity, the response to toxic substance, NADH dehydrogenase complex assembly, cytokine production, vesicle and vacuole organization. In turn, KEGG pathway tool classified DEGs that enrich molecular processes such as B and T cell receptor signaling pathways, natural killer cell-mediated cytotoxicity, Fc gamma R-mediated phagocytosis, toll-like receptor signaling pathways, TNF, mTOR signaling and Foxo signaling pathway, etc. Conclusions: In this descriptive study, our findings revealed the changes in the host transcriptome expression profile between HPL and LPL goats during SRLV infection and suggested that changes in proviral load induced altering the host’s metabolic network and expression of genes related to host immune responses, including inflammation, cell locomotion, cytokine production, defense response etc. The above findings provided unique insights for further studies on the mechanisms underlying SRLV-host interactions.

Project description:This investigation reports a differential proteomic analysis of the goat milk to evaluate and understand the protein changes induced by Staphylococcus spp. during a subclinical intramammary infection (IMI). A number of 9 milk samples from multiparous goats were selected for the study, 3 of which in mid-lactation (30-60 Days in milk - DIM) with very low somatic cell count (SCC) from half-udders producing a sterile milk bacterial culture (MLU), 3 of which in late lactation samples (> 250 DIM) with SCC > 2,000,000 cells/mL from half-udders producing a sterile milk bacterial culture for the whole lactation (LHU) and in conclusion 3 in late lactation (> 250 DIM) with SCC > 2,000,000 cells/mL from goat half-udders with a milk bacterial culture repeatedly positive for Staphylococcus aureus (LHS). Samples were analyzed using a shotgun proteomics approach, based on filter-aided sample preparation (FASP) followed by LC-MS/MS and on differential analysis conducted by spectral count approach (1).
