Project description:Constructed wetlands have been recognized as a removal treatment option for high concentrations of contaminants in agricultural waste before land application. The goal of this study was to characterize microbial composition in two constructed wetlands designed to remove contaminants from dairy washwater. Water samples were collected weekly for 11 months from two wetlands to determine the efficiency of the treatment system in removal of chemical contaminants and total and fecal coliforms. The reduction by the treatment was greatest for biological oxygen demand, suspended solids, chemical oxygen demand, nitrate, and coliforms. There was only moderate removal of total nitrogen and phosphorus. Changes in the total bacterial community and ammonia-oxidizing bacterial composition were examined by using denaturing gradient gel electrophoresis (DGGE) and sequencing of PCR-amplified fragments of the gene carrying the alpha subunit of the ammonia monooxygenase gene (amoA) recovered from soil samples and DGGE bands. DGGE analysis of wetlands and manure samples revealed that the total bacterial community composition was dominated by bacteria from phylogenetic clusters related to Bacillus, Clostridium, Mycoplasma, Eubacterium, and Proteobacteria originally retrieved from the gastrointestinal tracts of mammals. The population of ammonia-oxidizing bacteria showed a higher percentage of Nitrosospira-like sequences from the wetland samples, while a higher percentage of Nitrosomonas-like sequences from manure, feces, raw washwater, and facultative pond was found. These results show that the wetland system is a natural process dependent upon the development of healthy microbial communities for optimal wastewater treatment.

Project description:The microbial community diversity in Constructed Wetland System (CWS) plays a key role in the removal of pollutants from waste water. An integrated functional CWS developed at Neela Hauz Biodiversity Park, Delhi was selected to assess the diversity in composition and structure of microbial community diversity of sludge and sediment of CWS, based on metagenomic approach using 16S rRNA genes. The sediment showed higher diversity than sludge and both formed distinct clusters. The taxonomic structure of the microbial community of CWS is represented by 6,731 OTUs distributed among 2 kingdoms, 103 phyla, 227 classes, 337 orders, 320 families, 295 identified genera, and 84 identified species. The relative abundance of top 5 dominant phyla of sludge and sediment varied from 3.77% (Acidobacteria) to 35.33% (Proteobacteria) and 4.07% (Firmicutes) to 28.20% (Proteobacteria), respectively. The range of variation in relative abundance of top 5 dominant genera of sludge and sediment was 2.58% (Hyphomicrobium) to 6.61% (Planctomyces) and 2.47% (Clostridium) to 4.22% (Syntrophobacter), respectively. The rich microbial diversity of CWS makes it perform better in pollutants removal (59.91-95.76%) than other CWs. Based on the abundance values of taxa, the taxa are grouped under four frequency distribution classes-abundant (>20), common (10-19), rare (5-9), and very rare (1-4). The unique structure of microbial communities of integrated CWS is that the number of abundant taxa decreases in descending order of taxonomic hierarchy, while the number of rare and very rare taxa increases. For example, the number of abundant phyla was 14 and 21 in sludge and sediment, respectively and both communities have only 3 abundant genera each. This is in contrast to 4 and 17 very rare phyla in sludge and sediment, respectively and both the communities have 114 and 91 very rare genera, respectively. The outcomes of the study is that the integrated CWS has much higher microbial community diversity than the diversity reported for other CWs, and the rich diversity can be used for optimizing the performance efficiency of CWS in the removal of pollutants from waste water. Such structural diversity might be an adaptation to heterogeneous environment of CWS.

Project description:Conventional oil sewage treatment methods can achieve satisfactory removal efficiency, but energy consumption problems during the process of oil sewage treatment are worth attention. The integration of a constructed wetland reactor and a microbial fuel cell reactor (CW-MFC) to treat oil-contaminated wastewater, compared with a microbial fuel cell reactor (MFC) alone and a constructed wetland reactor (CW) alone, was explored in this research. Performances of the three reactors including chemical oxygen demand (COD), oil removal, and output voltage generation were continuously monitored. The COD removals of three reactors were between 73% and 75%, and oil removals were over 95.7%. Compared with MFC, the CW-MFC with a MnO₂ modified cathode produced higher power density and output voltage. Maximum power densities of CW-MFC and MFC were 3868 mW/m³ (102 mW/m²) and 3044 mW/m³ (80 mW/m²), respectively. The plants in CW-MFC play a positive role for reactor cathode potential. Both plants and cathode modification can improve reactor performance of electricity generation.

Project description:The objective of this study is to assess bioelectricity generation, pollutant removal (COD, ammonium, nitrate) and the bacterial communities on anodes in constructed wetlands coupled with microbial fuel cells (CW-MFCs), through feeding the systems with three different types of synthetic wastewater (system 1: normal wastewater; system 2: ammonium-free wastewater; system 3: nitrate-free wastewater). Three CW-MFCs were operated with different wastewater concentrations and hydraulic retention times (HRTs) over a long time period (6 months). The results indicate that the maximum open circuit voltage (775.63 mV) and maximum power density (0.628 W m-3) were observed in system 3 (period 3), and that bioenergy production was inhibited in system 2, when feeding with ammonium-free wastewater continuously. COD removal rates in the three systems were similar during each period and ranged from 82.2 ± 6.8% to 98.3 ± 2.2%. Ammonium removal occurred at the air cathode of the CW-MFCs through nitrification, and a higher level of ammonium removal was found in system 1 (period 3) compared with the others. Meanwhile, denitrification occurred at the anaerobic anode of the CW-MFCs, and a large amount of nitrate was removed effectively. The highest nitrate removal rate was 98.8 ± 0.5% in system 2 (period 3). Additionally, four genera related to electricity generation were detected at the anode: Geothrix; Desulfovibrio; Desulfobulbus; and Geobacter. The relative abundances of Desulfovibrio, Desulfobulbus and Geothrix gradually increased during the three periods in system 3, which might be beneficial for bioelectricity generation. Further investigations are needed to optimize the CW-MFC performance and explain the mechanism behind the pollutant degradation and electron motion in the CW-MFCs.
