Project description:Environmental temperature directly affects the concentration of chemicals in the gas phase. Therefore, if the olfactory system does not adapt to environmental conditions may provide inadequate information about distance to or direction of odor sources. Previous reports in Drosophila melanogaster show that temperature induces changes in olfactory sensitivity in complete individuals. Electrophysiological analysis demonstrates that these changes initiate at the olfactory receptor organs. In this experiment we try to identify particular genes responsible for olfactory adaptation to increasing temperatures using receptor organs of Drosophila after exposure to high temperatures. With this aim, wild-type Canton-S flies were subjected to 48-hour treatments at 30 degrees C. We use a direct approach selecting the specific tissue and making 'a priori' the gene election for the microarray analysis. Differential gene-expression analyses at the third antennal segments are performed comparing control and treated flies.

Project description:Olfaction and chemical communication are fundamental for coordination in insect societies, especially in species forming large colonies. Reproductive and task partitioning suggest that colony members belonging to different castes or subcastes or performing different tasks during their life (polyethism) may produce different semiochemicals and be differently sensitive to the variety of pheromones and/or environmental odours. The main peripheral olfactory organs are the antennal chemosensilla, where the early olfactory processes take place. At this stage, members of two different families of soluble chemosensory proteins (Odorant Binding Proteins -OBPs-, Chemosensory Proteins -CSPs-) show a remarkable affinity for different odorants and act as carriers while a further family, the Niemann Pick type C2 proteins (NPC2) may have a similar function, although this has not been fully demonstrated. Sensillar lymph also contains Odorant degrading enzymes (ODEs) which are involved in inactivation through degradation of the chemical signals, once the message is conveyed. Despite their importance in chemical communication, poor is known about how proteins involved in peripheral olfaction and, more generally antennal proteins, differ in honeybees of different caste, task and age. Here we investigate for the first time, using a shotgun proteomic approach, the antennal profile of honeybee of different castes (queens and workers) and of workers performing different tasks (nursers, guards and foragers) by controlling for the potential confounding effect of age. Regarding olfactory proteins, major differences were observed between queens and workers, some of which were found to be more abundant in queens (OBP3, OBP18, NPC2-1) and others to be more abundant in workers (OBP15, OBP21, CSP1, CSP3); while between workers performing different tasks, OBP14 was more abundant in nurses with respect to guards and foragers, and two ODEs were more abundant in guards with respect to workers of 2nd week. Apart from proteins involved in olfaction, we have found that the antennal proteomes are mainly characterized by castes and tasks, while age has a limited effect on antennal protein profile.

Project description:The goal of this study was to identify the preponderance of developmental hotspots in the Drosophila olfactory system. Using RNASeq-derived transcriptome profiles of the developing and adult antennae for six different Drosophila species, we show that a few highly variable transcription factors may drive, likely in a combinatorial fashion, high variability in a few select olfactory receptor neuron (ORN) developmental lineages (i.e. developmental hotspots) in an otherwise mostly conserved background. Furthermore, the high variability of these few ORN lineages leads to a correspondingly high probability that they will recruited during the adaptation of the Drosophila olfactory system to specific ecological pressures, one potential example being the remarkably convergent antennal transcriptome profiles in two Drosophila species that have independently evolved the ability to specialize on specific host plants.

Project description:To allow estimation of the complexity and gene expression differences of the antennal transcriptome between sexes as well as between castes, microarrays were designed based on an assembly of A. vollenweideri antennal sequence data from all sexes and castes. The microarrays were hybridized with samples generated from the respecitve antennal tissues, with four independent sample pools per sex and caste.

Project description:Allogrooming is a behaviour in which a worker uses its mouth parts to remove debris from the body of other colony members. This behaviour, observed in several species of eusocial insects, play a role in defence against parasites and pathogens. In honeybees, allogrooming represents an important resistance mechanism that seems to limit ectoparasites load, especially mites, within colonies and its expression depends on genetic and environmental factors. The degree of specialization of individuals performing this task is still unclear and behavioural and physiological correlates of allogroomers are largely unknown. It would be advantageous for allogroomers to be able to detect nestmates needing to be groomed. Since many stressors, including pathogens and parasites, change the odour of workers in A. mellifera, an enhanced perception of such chemical cues could contribute to a possibly specialized phenotype of allogroomers. Indeed, antennae play a key role in the expression of hygienic behaviours and proteomic investigation also showed that in the honeybee individuals performing different tasks differ for antennal profile of soluble olfactory proteins, which play a crucial role in the first steps of odour recognition. We thus investigated the expression of olfactory proteins in the antennae, predicting that if allogroomers have any degree of chemosensory specialization, this would reflect in a different antennal proteomic profile.
