Project description:Gata6 regulates lung epithelial stem cell development and airway regeneration. Here, the expression profile of microRNA was investigated when Gata6 was depleted during lung development. Analysis used total RNA from Sftpc-rtTA:tetO-cre lungs at E14.5 as control samples (WT) for comparison to the experimental samples of total RNA from Gata6flox/flox: Sftpc-rtTA: tetO-Cre mutant lungs at E14.5.

Single Cell Analysis of Mouse Lung Epcam+CD31/34/45- Epithelial Cells

Project description:Epithelial cell homeostasis and renewal in the lung requires the supportive signals from surrounding mesenchymal cells. Single cell RNA-seq analysis was used to examine the gene expression levels of several receptors on the Sftpc+ alveolar type II stem cells. Single cell RNA-seq revealed Ghr, Cxcr2, Cxcr4, and Igf1r expressing cells were all Sftpc+ cells in the normal mouse lung.

Expression of a surfactant protein C mutation impairs postnatal expansion of alveolar type 2 epithelial cells leading to permanent cell loss and decreased repair capacity in adult lungs

Project description:Mutations in the gene encoding surfactant protein C (SFTPC) are associated with interstitial lung disease in children and adults. To assess the natural history of disease, we knocked-in a familial, disease-associated SFTPC mutation, L188Q (L184Q in mice), into the mouse Sftpc locus. Expression of the L184Q allele (LQ) resulted in formation of an unstable, misfolded proprotein (proSP-CLQ) that was rapidly degraded by the proteasome pathway. Translation of proSP-CLQ exceeded that of proSP-CWT in neonatal AT2 cells and was associated with transient activation of oxidative stress and apoptosis leading to impaired expansion of AT2 cells during postnatal alveolarization. Consequently, adult mutant mice demonstrated a decrease in the number of AT2 cells with no effect on lung homeostasis; however, lung regeneration in response to bleomycin challenge was substantially reduced in mutant mice. Collectively, these data support the hypothesis that susceptibility to disease in adult mutant mice is established during postnatal lung development and is associated with a permanent reduction in AT2 cell numbers.

miRNAs regulate lung development and surfactant homeostasis via directly targeting SFTPC and TTF-1 in reprogramming-derived bovine model of NRDS

Project description:In this study, We proposed a reprogramming-derived bovine model of NRDS based on typical phenotypes of neonatal respiratory distress syndrome (NRDS) and high reproducibility and replication rates as well as a steerable background of neonatal cloned cows suffering from NRDS. With the aid of miRNA sequencing, construction of the miRNA–gene network and functional enrichment of target genes, the effects of specific miRNAs on lung development and surfactant homeostasis were determined in this model. We identified 12 miRNAs targeting SFTPB, SFTPC and NKX2-1 genes, demonstrated repression effects of three miRNAs on NKX2-1 and SFTPC expression in vivo and through which miRNAs disturbed lung development and surfactant homeostasis in the NRDS bovine model.

Transcription profiling of human HEK293 cells transfected with either wild type or delta exon 4 mutants of surfacant protein C

Project description:Mutations in the gene encoding surfactant protein C (SFTPC) have been linked to interstitial lung disease in children and adults. Expression of the index mutation, SP-Cdeltaexon4, in transiently transfected cells and type II cells of transgenic mice resulted in misfolding of the proprotein, activation of ER stress pathways and cytotoxicity. In the current study we show that stably transfected cells adapted to chronic ER stress imposed by constitutive expression of SP-Cdeltaexon4 via an NF-kB-dependent pathway. However, infection of cells expressing SP-Cdeltaexon4 with respiratory syncytial virus resulted in significantly enhanced cytotoxicity associated with accumulation of the mutant proprotein, pronounced activation of the unfolded protein response and cell death. Adaptation to chronic ER stress imposed by misfolded SP-C was associated with increased susceptibility to viral-induced cell death. The wide variability in the age of onset of ILD in patients with SFTPC mutations may be related to exposure to an environmental insult that ultimately overwhelms the homeostatic, cytoprotective response.
