Project description:In the title mol-ecule, C(16)H(15)N(3), the indole ring system is planar within 0.021 (3) Å and the phenyl ring is inclined to this plane by 17.32 (14)°. π-π contacts involving the pyrrole rings of inversion-related indole units [centroid-centroid distance = 3.5187 (17) Å] stabilize the crystal structure.

PCW-1001, a novel pyrazole compound, modulates the expression of DNA damage response genes

Project description:As pyrazole and its derivatives have a wide range of biological activities, including anticancer activity, the design of novel pyrazole derivatives has emerged as an important research field. This study describes a novel pyrazole derivative that exerts antitumor and radiosensitizing activities in breast cancer both in vitro and in vivo. We synthesized a novel pyrazole compound N,N-dimethyl-N'-(3-(1-(4-(trifluoromethyl)phenyl)-1H-pyrazol-4-yl)phenyl)azanesulfonamide (PCW-1001) and showed that it inhibited several oncogenic properties of breast cancer both in vitro and in vivo. PCW-1001 induced apoptosis in several breast cancer cell lines. Transcriptome analysis of PCW-1001-treated cells showed that it regulated the DNA damage response, suggesting its potential use in radiotherapy. Indeed, PCW-1001 enhanced the radiation sensitivity of breast cancer cells by modulating the expression of DNA damage response genes. Therefore, our data describe a novel pyrazole compound, PCW-1001, with antitumor and radiosensitizer activities in breast cancer.

The imidazole ring makes dihedral angles of 67.46?(10) and 23.10?(11)° with the attached benzene and phenyl rings, respectively. The dihedral angle between the benzene and phenyl rings is 68.22?(10)°. Inter-molecular O-H?N and C-H?N hydrogen bonds are found in the crystal structure.

Project description:The mol-ecular conformation of the title compound, C(20)H(17)N(3), is stabilized by an intramolecular C-H?N inter-action. The crystal structure shows inter-molecular C-H?? inter-actions. The dihedral angle between the isoquinoline unit and the phenyl ring is 11.42?(1)° whereas the isoquinoline unit and the pendent dimethyl pryrazole unit form a dihedral angle of 50.1?(4)°. Furthermore, the angle between the mean plane of the phenyl ring and the dimethyl pyrazole unit is 47.3?(6)°.

Project description:The title Schiff base compound, C(23)H(23)N(5)O, was synthesized by the reaction of 4-amino-phenazone and 3,5-dimethyl-1-phenyl-pyrazole-4-carbaxaldehyde. The mol-ecule adopts an E configuration about the central C=N double bond. A weak intra-molecular C-H?O hydrogen bond generates an S(6) ring motif. The dihedral angle between the pyrazole rings is 24.72?(10)° and the dihedral angles between the pyrazole rings and the adjacent phenyl rings are 58.67?(10) and 46.58?(11)°. The crystal structure is stabilized by weak C-H?? inter-actions involving the pyrazolone and phenyl rings.

The crystal structure is composed of two mol-ecules, 2-n-butyl-6-chloro-1-(2,4-dimethylphenyl-sulfon-yl)-1H-benzimidazole and 1-(2,4-dimethylphenyl-sulfon-yl)-2-n-butyl-5-chloro-1H-benz-imidazole, in the refined ratio of 0.759 (4):0.241 (4) disordered at the same position in the unit cell. The mol-ecule has three essentially planar fragments viz. benzimidazole, dimethyl-benzene and n-butyl (r.m.s. deviations of 0.009, 0.024 and 0.003 Å, respectively). The angle between the benzimidazole and dimethyl-benzene fragments is 86.0 (1)°. In the crystal, pairs of inter-molecular C-H⋯π inter-actions form centrosymmetrical dimers, which are linked by weak inter-molecular C-H⋯O hydrogen bonds.
