Project description:To determine differential expression of microRNAs in placentae with severe pre-eclampsia and normal placenta. Differential expression of microRNAs in placentae (4 severe pre-eclampsia and 4 normal control) was screened by microarray platform, then some differential microRNAs were selected and validated with real-time quantitative reverse transcription polymerase chain reaction in placentae of severe pre-eclampsia (n=24) and normal control group (n=26). Results: We validated the expression of some microRNAs altered in the microarray, and found the following microRNAs were significantly increased in severe pre-eclampsia placentae: miR-16, miR-29b, miR-195, miR-26b, miR-181a, miR-335 and miR-222. Conclusion: These differential microRNAs may play an important role in pathogenesis of pre-eclampsia.

Project description:Background: Preeclampsia, a pregnancy complication with hypertension and proteinuria, seriously threats the health and lives of the mother and the baby. The role of peripheral natural killer cells (NK cells) in the pre-eclampsia is unclear. Methods: Flow cytometry was performed to detect the expression of CD158a (KIR2DL1) and CD158b (KIR2DL2/3) in peripheral NK cells of non-pregnant healthy women (HW), healthy pregnant women (HP), and patients with pre-eclampsia (PE). Differentially expressed genes (DEGs) in CD158a+ and CD158b+ NK cells were identified by RNA-sequencing and real-time PCR. Protein array analysis was used to identify altered protein levels in the serum of study participants. Findings: CD158a+ NK cell numbers were significantly increased in the peripheral blood of patients with pre-eclampsia while the number of CD158b+ NK cells was significantly reduced. The percentage of CD158a+ NK cells was positively correlated with systolic blood pressure while the percentage of CD158b+ NK cells was negatively correlated with systolic blood pressure. RNA-seq and real-time PCR showed that the expression of ERAP2 and GCH1, the genes that regulate blood pressure and angiogenesis, was decreased in CD158a+ compared to CD158b+ NK cells. Interpretation: CD158a+ and CD158b+ NK cell numbers are altered in the peripheral blood of patients with pre-eclampsia compared to healthy individuals, which is associated with the increase in blood pressure.

Project description:The etiology of preeclampsia, a hypertensive disorder of human pregnancy, remains unknown. We addressed fetal sex selection and the suggestive role of fetal HLA-G and related genes, regulating maternal immune responses, in preeclampsia pathogenesis. We assessed birth sex ratios, weights, and seasonality of preeclampsia among 1.79 million births in Finland. We studied haplotypes of HLA-G 3’ untranslated region (UTR), regulating HLA-G expression, in 1000 Genomes series and in a preeclampsia cohort (n=1249). We quantified placental (n=163) mRNA expression of 136 genes, studied HLA-G and IFNα protein expression by immunohistochemistry, and measured maternal and fetal circulating IFNα levels by ELISA. Population-level data showed loss of male fetuses as a characteristic of preeclampsia. As a potential contributor to immune-mediated loss, we found balancing selection at HLA-G 3’UTR modulating sex ratio, and association of HLA-G 3’UTR haplotypes with placental HLA-G expression. HLA-G and its receptors were downregulated in preeclampsia placentas, and surprisingly, interferon alpha-1 (IFNA1) was highly upregulated. IFNA1 and HLA-G distinguished preeclampsia better than placental FLT1 expression. Fetal but not maternal circulating IFNα, produced by trophoblasts, showed association with maternal hypertension and fetal growth restriction. We uncover the link between placental HLA-G expression and human birth sex ratio. We propose that preeclampsia shares, through reduced HLA-G mediated immunotolerance, the mechanism needed to fight placental viral infections and malaria in evolution. IFNα upregulation in preeclampsia placenta, together with its known actions upstream of inflammatory genes, encourages testing IFNα inhibitors and especially the pregnancy-approved antimalarial hydroxichloroquine in treatment of preeclampsia.

Project description:The high incidence of pre-eclampsia, affecting 5-10% of pregnancies, makes it a major health problem. Early detection of pre-eclampsia, before the appearance of the first clinical symptoms, is therefore an urgent need, since it would allow the early intervention. Identification of plasma/serum biomarkers would pave the way to develop new strategies for the diagnosis of pre-eclampsia. Liquid biopsy emerges as a promising source of protein biomarkers that circumvent some of the inherent challenges of proteome wide analysis of plasma and serum. Besides, purified exosomes have the added interest of being communication vehicles between cells and tissues both in physiological and pathological processes. Methods We compared the protein abundance in purified exosomes from three different cohorts of control and preeclamptic serum samples, obtained around the sixth month and at the end of pregnancy. To this end, a shotgun label-free proteomics analysis was conducted and the relevant differential proteins were then validated by targeted MRM. Results An exosome purification method was developed that yielded highly enriched preparations, as indicated by the detection of a large number of exosomal markers. Likewise, the presence of specific pregnancy protein markers suggested that a very significant percentage of purified exosomes come from tissues related to pregnancy. Shotgun quantitative proteomic analysis allowed us to identify 10, 114 and 98 differentially regulated proteins in the three studied cohorts, with a high degree of concordance among them. Functional analysis suggests that these proteins participate in biological processes closely related to pre-eclampsia, such as angiogenesis, inflammation or cell migration. Differential abundance of 66 proteins were validated by MRM. Finally, we show the impact of the pre-eclampsia associated exosomes in the proteome of a cellular model, compared to normal exosomes. Conclusions We identified and validated differential proteins in liquid biopsy of patients that open new possibilities for the early diagnostic of pre-eclampsia. Additionally, the functional impact of the different proteome composition of purified pre-eclamptic exosomes in target cells provide new information to further understand changes on embryo-maternal interactions and therefore the pathogenesis of this disease.
