RNA-seq analysis of the first mandibular molars from K14-Cre; Wnt10aflox/flox mice and littermate control

Project description:Wnt ligands are continuously expressed during tooth development and are essential for cell proliferation; therefore, we hypothesized that other Wnt ligands may be up-regulated to compensate the loss of Wnt10a. To test this hypothesis, we investigated mRNA expression levels in the first mandibular molars at PN4 by RNA-seq analysis. A list of 26 Wnt pathway associated genes were identified to be expressed differently in K14-Cre;Wnt10aflox/flox mice. Among them, 7 genes were up-regulated and 19 genes were down-regulated. And the up-regulation of Wnt4 appeared to be the most significant. Moreover, heat map displayed that among the Wnt ligands with relatively high endogenous expression level, Wnt4 was up-regulated in K14-Cre;Wnt10aflox/flox mice than in control.

Increased gene expression in the peridontium of unopposed molars in rats

Project description:The crowns of the right maxillary molars were removed in 28 days old male Wistar rats (experimental). Gene expression patterns were examined in the periodontal ligaments (PDLs) of the ipsilateral and contralateral mandibular molars, three and fifteen days later. Rats whith no dental interventions served as controls. We used microarrays to detail gene expression underlying post-emergent tooth eruption and identified distinct classes of up-regulated genes during this process.

Comparative transcriptome profiles of human dental pulp cells from maxillary and mandibular teeth

Project description:PITX1 had a significantly higher expression in the lower teeth compared to the upper teeth and this difference in PITX1 level was more evident in the molars compared to premolars, consistent with data in mouse developing teeth. These show that the differential gene expression during odontogenesis can continue to exist in mature teeth. We suggest that an in vitro study of dental pulp cells should take the differential gene profiles between the mandibular and maxillary teeth into consideration. The knowledge about gene profiling and pathways in mature teeth paves a way to explore a more precise treatment approach in regenerative dentistry.

Project description:Single nucleotide polymorphisms in intron 1 of the fat mass and obesity-associated (FTO) gene were found to be associated with an increased risk of adult obesity. Enhanced FTO expression in mice leads to hyperphagia, increased fat mass, and higher body weight. Neuronal-specific FTOâ??deleted mice have an identical lean body weight phenotype to global FTO-deleted mice. The physiological role of adipose FTO in the homeostasis of energy regulation remains to be elucidated. We used microarrays to elucidate the metabolic pathways that are regulated by FTO in the white fat. FTO flox/flox and Adiponectin-cre FTO flox/flox (AFO) mice were fed with chow diet. White fat tissues from epididymal adipose pad were harvested under ad lib condition for RNA isolation. Three independent pools of FTO flox/flox and AFO mouse white fat RNA were included in the study.
