Project description:The interactions of various acyclic and cyclic hydrocarbons in both saturated and unsaturated forms with the carbon nanostructures (CNSs) have been explored by using density functional theory (DFT) calculations. Model systems representing armchair and zigzag carbon nanotubes (CNTs) and graphene have been considered to investigate the effect of chirality and curvature of the CNSs toward these interactions. Results of this study reveal contrasting binding nature of the acyclic and cyclic hydrocarbons toward CNSs. While the saturated molecules show stronger binding affinity in acyclic hydrocarbons; the unsaturated molecules exhibit higher binding affinity in cyclic hydrocarbons. In addition, acyclic hydrocarbons exhibit stronger binding affinity toward the CNSs when compared to their corresponding cyclic counterparts. The computed results excellently corroborate the experimental observations. The interaction of hydrocarbons with graphene is more favorable when compared with CNTs. Bader's theory of atoms in molecules has been invoked to characterize the noncovalent interactions of saturated and unsaturated hydrocarbons. Our results are expected to provide useful insights toward the development of rational strategies for designing complexes with desired noncovalent interaction involving CNSs.

Project description:Dietary unsaturated fatty acids beneficially affect human health, in part by modulating the immune system, but the mechanism is not completely understood. Given that unsaturated fatty acids have been shown to be covalently incorporated into a small subset of proteins, we designed three alkyne-tagged chemical reporters of unsaturated fatty acids, alk-16:1, alk-17:1 and alk-18:1, to explore the generality and diversity of this protein modification. Following cell lysis, proteins labelled with the reporters could be captured by azido-functionalized reagents via CuAAC for fluorescence detection or enrichment for proteomics analysis. These reporters label many proteins in mammalian cells and can be incorporated site-specifically, notably on Cys residues. Quantitative proteomics analysis (n= 4 biological replicates) of LPS/IFN-gamma stimulated RAW264.7 labelled with oleic acid (control), alk-16 (palmitic acid chemical reporter), alk-16:1, alk-17:1 and alk-18:1, revealed that unsaturated fatty acids modify similar protein targets to saturated fatty acids, including several immune proteins. Interestingly, some proteins can be differentially labeled with unsaturated and saturated fatty acid.

Project description:We present experimental measurements and analysis of the dynamics and the phase behaviour of saturated DMPC and unsaturated DOPC oriented multi-lamellar bilayers. Elastic and inelastic neutron scattering were used to directly probe the dynamical processes of these membrane systems on time and length scales relevant to the internal and localized motion of lipid monomers. Mobility in this regime can be informative in elucidating the local interactions responsible for material properties of these fluid lipid systems. DMPC and DOPC are structurally similar in terms of their membrane hydrophobic thickness; however, they exhibit different mechanical properties in terms of both elastic compressibility and bending moduli. The analyses suggest that the constraint imposed by the double bonds in DOPC acyl chains restricts atomic motion in both liquid and gel phases compared to DMPC. We discuss applications of molecular dynamics to further elucidate the atomic details of the dynamical processes. Such an understanding may suggest how membrane properties can be tuned using a variety of different lipid species.

Project description:The objective of this research was to investigate the retention and transport behavior of GenX in five natural porous media with similar median grain diameters but different geochemical properties. Surface tensions were measured to characterize surface activity. Miscible-displacement experiments were conducted under saturated conditions to characterize the magnitude of solid-phase adsorption, while unsaturated-flow experiments were conducted to examine the impact of air-water interfacial adsorption on retention and transport. The results from surface-tension measurements showed that the impact of solution composition is greater for the ammonium form of GenX than for the acid form, due to the presence of the NH4 counterion. The breakthrough curves for the experiments conducted under saturated conditions were asymmetrical, and a solute-transport model employing a two-domain representation of nonlinear, rate-limited sorption provided reasonable simulations of the measured data. The magnitudes of solid-phase adsorption were relatively small, with the highest adsorption associated with the medium containing the greatest amount of metal oxides. The breakthrough curves for the experiments conducted under unsaturated conditions exhibited greater retardation due to the impact of adsorption at the air-water interface. The contributions of air-water interfacial adsorption to GenX retention ranged from ∼24% to ∼100%. The overall magnitudes of retardation were relatively low, with retardation factors < ∼3, indicating that GenX has significant migration potential in soil and the vadose zone. To our knowledge, the results presented herein represent the first reported data for solid-water and air-water interfacial adsorption of GenX by soil. These data should prove useful for assessing the transport and fate behavior of GenX in soil and groundwater.

Project description:The aim of this study was to determine the effects of unprotected dietary unsaturated fatty acids (UFA) from different plant oils on gene expression in the mammary gland of grazing dairy cows. Milk composition and gene expression in the mammary gland tissue were evaluated in grazing dairy cows supplemented with different unsaturated fatty acids (UFA). The UFA supplementation improves the health and nutrition quality aspects of dairy milk, but also affects the gene networks expression signature associated with cellular growth and proliferation, cell-death, signalling, nutrient metabolism, and immune response, and in turn, the mammary gland integrity and health. Project description:The study aimed to identify the lateral heterogeneity of soil physicochemical properties in riparian zones, and its underlying drivers during natural restoration after agricultural abandonment. Abandoned farmlands, after 5-year natural restoration, within 500?m from the edges on both sides of Liaohe River were selected as the study area. Soil physicochemical properties of four lateral buffers (<10?m, 10~100?m, 100~300?m, and?>300?m from river edge, respectively) along riparian zones were measured. The results showed that riparian soils were characterized by high sand content (78.88%~96.52%) and poor soil nutrients. Soil silt content, organic carbon (OC), cation exchange capacity (CEC), total nitrogen (TN), and available nitrogen (AN) increased laterally with increasing distance from river edge, while soil sand content decreased. Total phosphorus (TP) and available phosphorus (AP) are not spatially autocorrelated. Soil OC, TN, AN, and CEC along upstream and midstream reaches showed negative spatial autocorrelation along the lateral gradients, and positive along downstream reach. Altitude, distance from river edge and distance from nearest farmland were the pronounced factors affecting soil physicochemical properties in this study.

Project description:River ecosystems receive and process vast quantities of terrestrial organic carbon, the fate of which depends strongly on microbial activity. Variation in and controls of processing rates, however, are poorly characterized at the global scale. In response, we used a peer-sourced research network and a highly standardized carbon processing assay to conduct a global-scale field experiment in greater than 1000 river and riparian sites. We found that Earth's biomes have distinct carbon processing signatures. Slow processing is evident across latitudes, whereas rapid rates are restricted to lower latitudes. Both the mean rate and variability decline with latitude, suggesting temperature constraints toward the poles and greater roles for other environmental drivers (e.g., nutrient loading) toward the equator. These results and data set the stage for unprecedented "next-generation biomonitoring" by establishing baselines to help quantify environmental impacts to the functioning of ecosystems at a global scale.

Project description:While outdoor recreationists often report increases to their well-being for time spent in nature, the mechanisms through which local ecologies affect human health have been difficult to quantify, and thus to manage. We combine data from pre-post salivary cortisol measures, GPS tracks, visitor photos, and surveys from 88 hikers traversing several types of landscape within peri-urban public lands in southwest Idaho, USA. We find that time in biodiverse riparian areas and areas of perceived aesthetic value correlates with decreases in salivary cortisol and improved well-being for hikers. Wildlife sightings were not associated with changes in salivary cortisol, but were associated with riparian travel and aesthetic preferences, indicating an indirect pathway for ecosystem services. Additionally, wildlife sightings decreased on high-use days, even though hikers did not perceive a negative impact of their recreational activity. These results suggest that cultural and physiological ecosystem services of nature depend on the ecological community of the area. Preferential visitation and high service value of riparian areas by hikers and wildlife alike target shared riparian areas as hot spots for management efforts to promote both ecological and human health within an increasingly urbanizing world.
