While most tests have studied nanoparticle effects at organismal levels (e.g. mortality, reproduction), effects at suborganismal levels may clarify the role of metal ions, nanoparticles and nanoparticle aggregates. In this study, the effect of ZnO, CuO nanoparticles and zinc, copper salts was tested on the gene expression levels in Daphnia magna. D. magna was exposed during 96 hours to 10% immobilization concentrations of all chemicals, after which daphnids were sampled for a differential gene expression analysis using microarray. When comparing the nanoparticle exposed daphnids (ZnO or CuO) to the metal salt exposed daphnids (zinc or copper salt), the microarray results showed no significantly differentially expressed genes. Project description:Purpose: The intensive use of metal-based nanoparticles results in their continuous release into the environment and the subsequent destroying microbial biodiversity and causing occurrence of antibiotic resistant determinants. Although previous studies have indicated that nanoparticles may be toxic to microorganisms, there is a scarcity of data available to assess the underlying molecular mechanisms of inhibitory and biocidal effects of nanoparticles on microorganisms, which is a critical gap in our comprehensive understanding of the impacts of nanoparticles on microbial ecosystems. Methods: By combining the global activated/suppressed gene profiles and detected physiological responses, we detected the microbial response to CuO NPs exposure and revealed potential mechanisms of CuO NPs on this prevalent opportunistic pathogen and environmentally relevant denitrifier. Results:Our results indicate that nanoparticles could react directly with the biological membrane, but also change gene expressions. Transmission electron microscopy (TEM) results indicate that CuO nanoparticles not only damaged the structure of the bacterial membrane but also entered the cells. Planktonic cells of P. aeruginosa are energetically compromised after exposure to a sublethal level (1 and 10 mg/L) of CuO nanoparticles. Respiration was likely inhibited as denitrification activity was severely depleted in terms of decreased transcript levels of most denitrification genes. In addition, and of high concern, bacteriophage genes were activated and it is speculated that phage mediated cell lysis. Meanwhile, CuO NP exposure induced significantly up-regulated expressions of metal resistance gene, resistance-nodulation-division, P-type ATPase efflux and cation diffusion facilitator transporters, which can form an integrated network controlling metals concentrations in the cytoplasm and periplasm P. aeruginosa. Conclusion: Based on our results, it is evident that the response of bacteria under the exposure of nanoparticles is very complex, though a clear and comprehensive understanding of the true mechanisms of inhibition or toxicity is still lacking. Our findings will provide insights on whole picture regarding the fundamental mechanisms of microbial susceptibility, tolerance and resistance to exposure of CuO NPs, as well highlight to re-estimate potential risks of CuO NP to public health and environment.

Project description:The objective of the study was to investigate how pristine CuO modulates allergic lung inflammation and whether surface modifications can influence its reactivity. CuO materials exacerbated the existing allergic airway inflammation by eliciting dramatic pulmonary neutrophilia. Transcriptomic analysis showed that CuO, CuO COOH and CuO NH3 commonly enriched neutrophil-related biological processes in lungs with and without the pre-existing condition. In contrast, CuO PEG had a significantly lower potential in triggering pulmonary changes revealing that surface PEGylation suppresses the effects triggered by the pristine material.

Project description:Nanopesticides (NPe) are innovative pesticides involving engineered nanomaterials (ENM) to increase the efficiency of plant protection products. However, no toxicological data is available on their inhalation toxicity and the potential cocktail effects between pesticide active substances and ENM. In particular, the neurodevelopmental toxicity of prenatal repeated exposures has not been specifically evaluated. To this end, C57Bl/6j gestating mice were exposed to aerosols in an ad hoc whole-body exposure chamber. Exposures consisted in low repeated doses of the paraquat herbicide (PQ), and/or TiO2 nanoparticles (nTiO2). The monitoring of aerosol characteristics, in addition to their prior metrological characterization, enabled a precise estimation of the exposure doses received by animals, which can be extrapolated to the recommended exposure limit proposed by health agencies. We used RNA-seq to assess the gene expression in pups striatum at post-natal stage, in order to highlight possible neurodevelopmental alterations. This sensitive method revealed that both PQ and nTiO2 aerosols had a significant effect on the gene expression of the offspring, highlighting dysregulations of oligodendrocyte differentiation and neuronal maturation. Exposure to the nTiO2 aerosol modulated notably the regulation of neurotransmitters pathway being possibly indirect consequences of a minor change in the neuroinflammation state. No synergy was found between the PQ + nTiO2. Instead, the neurodevelopmental effects were surprisingly lower than the one measured for each substance separately.
