LongSAGE transcriptome comparison on Botrytis cinerea conidia germination with and without resveratrol treatment

Project description:Two long serial analysis of gene expression (LongSAGE) libraries of germinating conidia of Botrytis cinerea were generated with and without resveratrol treatments for unraveling the antifungal mechanisms of this phytoalexin by global transcriptome analysis – 82,786 total tags and a minimum of 15,665 unitags were obtained respectively. By tag-to-gene mapping, about 55% of the total unitags were matched to known genes. There were 109 unitags that showed significant differences in the two LongSAGE libraries, corresponding to 43 up- and 44 down-regulated genes, and 22 unmatched unitags as new gene candidates. These results showed that resveratrol inhibited expression of genes related to the glycolysis-Krebs cycle and mitochondrial electron transport chains at multiple sites in conidia germination, with induction of modified Entner–Doudoroff pathway as metabolic compensation. The chaperone-mediated protein folding and ubiquitin proteolysis systems were inhibited, while detoxification of resveratrol via ABC transporters was activated in this fungal pathogen. WRKY33 binding sites in Arabidopsis upon Botrytis cinerea 2100 inoculation

Project description:The Arabidopsis thaliana mutant wrky33 is highly susceptible to the necrotrophic fungus Botrytis cinerea. We identified by ChIP-seq >1680 high-confidence WRKY33 binding sites associated with 1576 genes within the Arabidopsis genome, with all of them being dependent on rapid activation of WRKY33 expression by Botrytis cinerea strain 2100. Genome-wide transcriptional analysis defined 318 genes as direct functional targets at 14 h post inoculation. Comparison between resistant wild-type Columbia-0 and susceptible wrky33 mutant plants revealed that expression of 75% of all WRKY33 regulated targets were down-regulated upon infection, indicating that WRKY33 predominately acts as a repressor. However, WRKY33 appears to possess dual functionality acting either as a repressor or as an activator in a promoter-context dependent manner. Our genome-wide analysis confirmed known WRKY33 targets involved in ethylene and jasmonic acid hormone signaling and phytoalexin biosynthesis, but also uncovered a previously unknown role of abscisic acid (ABA) biosynthesis in the complex regulatory circuitry affecting resistance towards Botrytis. Transcription profiling of tomato fruit responses to Botrytis cinerea

Project description:Tomato fruit ripening is associated with a dramatic increase in susceptibility to the fungal pathogen Botrytis cinerea, the causal agent of gray mold. Mature green fruit, prior to ripening, are largely resistant to B. cinerea, whereas red fruit, at the end of ripening, are susceptible to B. cinerea infection. We used microarrays to detail the gene expression changes that are induced by B. cinerea when tomato fruit at unripe and ripe stages are infected. Experiment Overall Design: Tomato fruit at mature green and red ripe stages were wound inoculated with a water suspension of B. cinerea conidia. Twenty four hours post inoculation fruit pericarp and epicarp tissue around and including the inoculation sites was collected and the total RNA extracted. Total RNA was also collected from healthy and mock inoculated fruit.

Acetylome analysis of Botrytis cinerea

Project description:Lysine acetylation of proteins is a dynamic and reversible post-translational modification that plays an important role in diverse cellular processes. Botrytis cinerea is the most thoroughly studied necrotrophic species due to its broad host range and huge economic impact. However, to date, little is known about the functions of lysine acetylation in this plant pathogen. In this study, we determined the lysine acetylome of B. cinerea through the combination of affinity enrichment and high-resolution LC-MS/MS analysis. Overall, 1582 lysine acetylation sites in 954 proteins were identified.
