Project description:Thioguanine (6-TG) as conventional drug has anti-tumor activity in various cancer cell lines. However, the effect of 6-TG on luminal subtypes breast cancer (ER+, PR+) is elusive. In this study, effect of 6-TG on MCF-7 breast cancer cell line (ER+, PR+) and its mechanisms were investigated. MCF-7 cells were treated with 6-TG and IC50 value was measured by cell counting kit-8 assay. DEGs were confirmed by RNA-seq. Apoptosis and cell cycle consequences were checked by flow cytometry and western blot. Our results showed that colony formation decreased obviously after 6-TG treatment. Moreover, with the treatment of 6-TG cell apoptosis percentage increased. DNMT1 mRNA and protein expression decreased and FAS expression increased in both expression level. Meanwhile, 6-TG treatment also induced MCF-7 cells to occur G2/M phase cell cycle arrest. Overall, our results indicated that 6-TG suppressed cell proliferation, induced G2/M phase arrest through DNMT1 mediated p53-dependent p21 signaling, and accelerated cell apoptosis through activated extrinsic apoptosis pathway which was caused by DNMT1 mediated FAS signaling.

Project description:The DNA methyltransferase activity of DNMT1 is vital for genomic maintenance of DNA methylation. We report here that DNMT1 function is regulated by O-GlcNAcylation, a protein modification that is sensitive to glucose levels, and that elevated O-GlcNAcylation of DNMT1 from high glucose environment leads to alterations to the epigenome. Using mass spectrometry and complementary alanine mutation experiments, we identified S878 as the major residue that is O-GlcNAcylated on DNMT1. Functional studies further revealed that O-GlcNAcylation of DNMT1-S878 results in an inhibition of methyltransferase activity, resulting in a general loss of DNA methylation that is preferentially at partially methylated domains (PMDs). This loss of methylation corresponds with an increase in DNA damage and apoptosis. These results establish O-GlcNAcylation of DNMT1 as a mechanism through which the epigenome is regulated by glucose metabolism and implicates a role for glycosylation of DNMT1 in metabolic diseases characterized by hyperglycemia.

Project description:We report that Dnmt1 is crucial during perinatal intestinal development. Loss of Dnmt1 in intervillus progenitor cells causes global hypomethylation, DNA damage, premature differentiation, and apoptosis, and consequently, loss of nascent villi. We further confirm the critical role for Dnmt1 during crypt development using the in vitro organoid culture system, and illustrate a clear differential requirement for Dnmt1 in immature versus mature organoids. These results demonstrate an essential role for Dnmt1 in maintaining genomic stability during intestinal development and the establishment of intestinal crypts.

Project description:The combination of natural products with standard chemotherapeutic agents offers a promising strategy to enhance the efficacy of standard chemotherapy by reducing their dosage and side effects. The standard chemotherapeutic drug for breast and other cancer types- doxorubicin(DOX), also known as Adriamycin (an anthracycline) has several disadvantages including severe side effects and development of drug resistance. Recently, we reported the broad spectrum pharmacological activity and potential markers of Australian propolis extract (AP-1). In the present study, we explored the synergistic interactions of AP-1 with DOX against MCF-7 cells by implementing different synergy quantitation models. To identify the potential anticancer metabolites in AP-1, biochemometric and metabolomics-driven study was performed. Furthermore, we evaluated the molecular mechanisms involved by analysing the apoptotic profile (using flow cytometry and proteome array of 35 apoptotic proteins) and oxidative status (using reactive oxygen species detection) of the MCF-7 cells upon treatment with the most synergistic combination. The underlying synergistic mechanism against the MCF-7 cells was also studied using label-free quantification proteomics analysis. Five prenylated stilbenes were identified as the most discriminant metabolites in the most active AP-1 fraction, three of them previously isolated from AP-1 with IC50 values in the range 0.68-2.7 µM against the MCF-7 cells. Strong synergy was observed when AP-1 was combined with DOX in the ratio of 100:0.29 (w/w) as validated by different synergy quantitation models including combination index (CI), HSA, ZIP, LOEWE, and BLISS. AP-1 significantly enhanced the inhibitory effect of DOX (p < 0.05) against the MCF-7 cell proliferation in a dose-dependent manner with significant reduction of ROS production (p < 0.05; compared to DOX alone). AP-1 also enhanced the apoptotic effect of DOX significantly after 24 h of treatment with a shift of DOX-induced necrosis to apoptosis. Significant upregulation of catalase, HTRA2/Omi, FADD together with TRAIL-mediated apoptosis, DR5 and DR4 (p < 0.05) was observed which contributed to the anti-proliferative activity of AP-1 against the MCF-7 cells. The enhancement of pro-apoptotic p27, PON2 and catalase with reduction of anti-apoptotic XIAP, HSP60 and HIF-1α may be associated with the improved apoptosis in the MCF-7 cells observed in the Annexin V-7AAD flow cytometry analysis of the synergistic combination compared to the mono treatments. Furthermore, a significant increase of antioxidant proteins including catalase and PON2 was observed upon treatment with the synergistic combination which may be associated with the parallel enhancement of apoptosis. Shotgun proteomics in the synergistic combination-treated cells identified 21 significantly dysregulated proteins involving the TP53/ATM-regulated non-homologous end-joining pathway and double-strand breaks repairs, recruiting the overexpressed BRCA1 and curtailed RIF1 encoded proteins. The overexpression of UPF2 was noticed after the treatment with the synergistic combination which could assist to overcome doxorubicin resistance-associated long non-coding RNA and decline the metastasis of the MCF-7 cells. In summary, we propose a promising AP-1 and DOX synergistic combination against the MCF-7 cells and highlighted the possible synergistic mechanisms of action together with the key prenylated metabolites of AP-1. Further in vivo and clinical studies are warranted on this synergistic combination.

Project description:The emergence of anti-estrogen resistance in breast cancer is an important clinical phenomenon affecting long-term survival in this disease. Identifying factors that convey cell survival in this setting may guide improvements in treatment. Estrogen (E2) can induce apoptosis in breast cancer cells that have been selected for survival after E2 deprivation for long periods (MCF-7:5C cells), but the mechanisms underlying E2-induced stress in this setting have not been elucidated. Here, we report that the c-Src kinase functions as a key adapter protein for the estrogen receptor (ER, ESR1) in its activation of stress responses induced by E2 in MCF-7:5C cells. E2 elevated phosphorylation of c-Src, which was blocked by 4-hydroxytamoxifen (4-OHT), suggesting that E2 activated c-Src through the ER. We found that E2 activated the sensors of the unfolded protein response (UPR), IRE1? (ERN1) and PERK kinase (EIF2AK3), the latter of which phosphorylates eukaryotic translation initiation factor-2? (eIF2?). E2 also dramatically increased reactive oxygen species production and upregulated expression of heme oxygenase HO-1 (HMOX1), an indicator of oxidative stress, along with the central energy sensor kinase AMPK (PRKAA2). Pharmacologic or RNA interference-mediated inhibition of c-Src abolished the phosphorylation of eIF2? and AMPK, blocked E2-induced ROS production, and inhibited E2-induced apoptosis. Together, our results establish that c-Src kinase mediates stresses generated by E2 in long-term E2-deprived cells that trigger apoptosis. This work offers a mechanistic rationale for a new approach in the treatment of endocrine-resistant breast cancer. MCF-7:5C cells were treated with vehicle (0.1% EtOH) as control, E2 (10-9mol/L), 4-OHT (10-6mol/L), E2 (10-9mol/L) plus 4-OHT (10-6mol/L), PP2 (5x10-6mol/L), and E2 (10-9mol/L) plus PP2 (5x10-6mol/L) respectively for 72 hours.

Project description:Previous study demonstrated that HDAC3 has a critical role in MM proliferation; however, the underlying mechanism has not yet been elucidated. We identify that HDAC3 inhibition targets DNMT1 through dual regulations. We demonstrate that knockdown of DNMT1 leads to apoptosis and significant growth inhibition in myeloma cells. HDAC3 inhibition by gene silencing or HDAC3 selective inhibitor BG45 downregulates an oncoprotein c-Myc through its acetylation. c-Myc directly regulates DNMT1 expression at its enhancer region. Furthermore, HDAC3 directly regulates the stability of DNMT1 protein through its acetylation. Pharmaceutical inhibition of HDAC3 and DNMT1 synergistically induce MM growth inhibition in in vitro and in vivo settings. The goal of this analysis is to identify genes whose expression changes after shRNA-mediated knockdown of HDAC3 or DNMT1 using the human U133 plus 2.0 Affymetrix GeneChip in myeloma cell line (MM.1S).
