Project description:We demonstrate a new high-throughput model where rapidly proliferating and easily handled induced pluripotent stem cells enable generation of human prostate tissue in vivo and, for the first time, in vitro. Using a co-culture technique with urogenital sinus mesenchyme, we recapitulated the in situ prostate histology, including the stromal compartment and the full spectrum of epithelial differentiation.

Proteomics of prostate cancer serum and plasma using low and high throughput approaches

Project description:Mass spectrometry-based proteomics has been successfully used to characterize biofluids, most notably blood, for biomarker discovery or to evaluate patients’ underlying diseases. Despite immense progress, challenges such as blood’s large dynamic range or low throughput remain for those types of studies. In this work, we used cutting-edge proteomics technologies to construct labelled and label-free l workflows, capable of quantifying approximately 2,000 proteins in biofluids. With just 70µL of blood and a single depletion strategy, we conducted an analysis of a cohort comprising 64 individuals, comparing prostate cancer patients to healthy donors. The results revealed dozens of differentially expressed proteins in both plasma and serum. Notably, we identified the upregulation of Prostate Specific Antigen (PSA), a well-known biomarker for prostate cancer, in the cancer cohort, along with the discovery of potential novel markers. Most of the differentially regulated proteins were consistently quantified using both the lower-throughput and higher-throughput workflows. Our bioinformatics analysis of the prostate cancer cohort data suggests that some of the newly discovered proteins may be secreted differentially into the bloodstream, making them potential candidates for disease markers.

Propagation of human prostate tissue from induced pluripotent stem cells.

Project description:Primary culture of human prostate organoids and patient-derived xenografts is inefficient and has limited access to clinical tissues. This hampers their use for translational study to identify new treatments. To overcome this, we established a complementary approach where rapidly proliferating and easily handled induced pluripotent stem cells enabled the generation of human prostate tissue in vivo and in vitro. By using a coculture technique with inductive urogenital sinus mesenchyme, we comprehensively recapitulated in situ 3D prostate histology, and overcame limitations in the primary culture of human prostate stem, luminal and neuroendocrine cells, as well as the stromal microenvironment. This model now unlocks new opportunities to undertake translational studies of benign and malignant prostate disease.

Project description:MYCN amplification and overexpression are common in neuroendocrine prostate cancer (NEPC). However, the impact of aberrant N-Myc expression in prostate tumorigenesis and the cellular origin of NEPC have not been established. We define N-Myc and activated AKT1 as oncogenic components sufficient to transform human prostate epithelial cells to prostate adenocarcinoma and NEPC including the small cell prostate carcinoma (SCPC) variant with phenotypic and molecular features of aggressive, late-stage human disease. We directly show that prostate adenocarcinoma and NEPC can both arise from a common epithelial clone. Further, N-Myc is required for tumor maintenance and destabilization of N-Myc through Aurora A kinase inhibition reduces tumor burden. Our findings establish N-Myc as a driver of NEPC and a target for therapeutic intervention. Expression profiling by high throughput sequencing of experimentally generated human tumors with mixed NEPC and prostate adenocarcinoma. Gene expression analysis of laser capture microdissected NEPC and adenocarcinoma from three independent engineered human tumors of mixed NEPC and prostate adenocarcinoma phenotype.
