Project description:The sea cucumber Apostichopus japonicus is a deposit-feeder and vital for marine benthic ecosystems. Hypoxia lasting for several days can lead to massive mortality in A. japonicus. It is important to understand the molecular responses of A. japonicus when exposed to acute changes in DO concentration. In the present study, RNA-seq provided a general overview of the gene expression profiles of the respiratory tree of A. japonicus exposed to dissolved oxygen (DO) of 8 mg/L (DO8), 4 mg/L (DO4) and 2 mg/L (DO2) conditions.

Arabidopsis thaliana ambient temperature responsive lncRNAs.

Project description:BACKGROUND:Long non-coding RNAs (lncRNAs) have emerged as new class of regulatory molecules in animals where they regulate gene expression at transcriptional and post-transcriptional level. Recent studies also identified lncRNAs in plant genomes, revealing a new level of transcriptional complexity in plants. Thousands of lncRNAs have been predicted in the Arabidopsis thaliana genome, but only a few have been studied in depth. RESULTS:Here we report the identification of Arabidopsis lncRNAs that are expressed during the vegetative stage of development in either the shoot apical meristem or in leaves. We found that hundreds of lncRNAs are expressed in these tissues, of which 50 show differential expression upon an increase in ambient temperature. One of these lncRNAs, FLINC, is down-regulated at higher ambient temperature and affects ambient temperature-mediated flowering in Arabidopsis. CONCLUSION:A number of ambient temperature responsive lncRNAs were identified with potential roles in the regulation of temperature-dependent developmental changes, such as the transition from the vegetative to the reproductive (flowering) phase. The challenge for the future is to characterize the biological function and molecular mode of action of the large number of ambient temperature-regulated lncRNAs that have been identified in this study.

Transcriptome analysis of cucumber in response to high temperature stress

Project description:In order to find out the genes involved in the heat shock response of cucumber, the seedlings of '9930', a North China type cucumber variety, were treated with high temperature at 42℃, and the leaves of the seedlings were taken after 0, 3 and 6 hours of treatment for transcriptome sequencing to analyze the differential expression genes of cucumber in response to high temperature stress. The results showed that compared with the control (heat treatment at 0h), there were 6 082 genes differentially expressed after heat treatment for 3 h and 6 h, of which 3245 were up-regulated and 2 388 were down-regulated. The results of enrichment analysis of GO and KEGG showed that these differentially expressed genes were mainly involved in metabolic pathway and protein synthesis pathway. Among the differentially expressed genes, there are many Hsp family genes, including Hsp20, HSP70 and Hsp90, which are up-regulated by heat stress, indicating that these genes may play a positive regulatory role in heat shock response of cucumber. In addition, many transcription factors, including AP2, MYB, WRKY, bHLH and HSF, were also induced by heat.
