Project description:The purpose of this study is to investigate if acute systemic dehydration impacts the vocal folds at the mRNA level. Rabbits were used as animal model to enable a systematic and rigorous assessment of the pathobiology of vocal fold hydration since they can be manipulated in a physiologically realistic manner. We anticipate that the results of this study will help to elucidate the effects of altered hydration state in response to furosemide on vocal fold epithelium, lamina propria and muscle tissue at the transcriptome level. Together with cellular, structural, and functional techniques that are part of this project, we expect to provide validated scientific recommendations on the adverse effects of dehydration and the therapeutic benefits of hydration.

Project description:The objective of this study was to obtain a comprehensive proteome of euhydrated and systemically dehydrated rabbit vocal folds. We hypothesized that a mild-moderate systemic dehydration level would lead to changes in protein levels. New Zealand White rabbits were randomly assigned to euhydrated (control) or furosemide-dehydrated groups. Systemic dehydration was assessed by body weight loss and changes in blood markers. Vocal fold specimens were processed for proteomic analysis using liquid chromatography-tandem mass spectrometry. Systemic dehydration induced 5.5 percent body weight loss (range: 4.5 to 6.5 percent) and significant changes in blood analytes compared to controls. More than 1500 proteins were identified across all vocal fold samples, associated with a variety of cellular components and ubiquitous cell functions such as protein synthesis and degradation, and mitochondrial translation and respiration. The comparison analysis identified 27 proteins differentially expressed in the systemically dehydrated vocal folds. These proteins are mainly involved with mitochondrial translation and metabolism, which are downregulated in response to systemic dehydration. We identified a large group of proteins representing the rabbit vocal fold tissue. The data provide novel insights on the response of vocal folds to a mild-moderate level of systemic dehydration that involves the downregulation of mitochondrial metabolism, suggesting a biologic mechanism to prevent oxidative damage associated with furosemide-induced dehydration.

Project description:Vocal folds are a viscoelastic multilayered structure responsible for voice production. Vocal fold epithelial damage may weaken the protection of deeper layers of lamina propria and thyroarytenoid muscle and impair voice production. Systemic dehydration can adversely affect vocal function by creating suboptimal biomechanical conditions for vocal fold vibration. However, the molecular pathobiology of systemically dehydrated vocal folds is poorly understood. We used an in vivo rabbit model to investigate the complete gene expression profile of systemically dehydrated vocal folds. The RNA-Seq based transcriptome revealed 203 differentially expressed (DE) vocal fold genes due to systemic dehydration. Interestingly, function enrichment analysis showed downregulation of genes involved in cell adhesion, cell junction, inflammation, and upregulation of genes involved in cell proliferation. RT-qPCR validation was performed for a subset of DE genes and confirmed the downregulation of DSG1, CDH3, NECTIN1, SDC1, S100A9, SPINK5, ECM1, IL1A, and IL36A genes. In addition, the upregulation of the transcription factor NR4A3 gene involved in epithelial cell proliferation was validated. Taken together, these results suggest an alteration of the vocal fold epithelial barrier independent of inflammation, which could indicate a disruption and remodeling of the epithelial barrier integrity. This transcriptome provides a first global picture of the molecular changes in vocal fold tissue in response to systemic dehydration. The alterations observed at the transcriptional level help to understand the pathobiology of dehydration in voice function and highlight the benefits of hydration in voice therapy.

Project description:Older subjects are more prone to develop systemic dehydration. Systemic dehydration has implications for vocal fold biology by affecting gene and protein expression. The interaction of hydration status and age has not been investigated in the context of vocal fold biology. We used comparative proteomics to analyze the vocal fold proteome of 6.5-month-old and over 3-year-old rabbits under water ad libitum or water volume restriction protocol.
Young and old rabbits (n= 22) were either euhydrated (water ad libitum) or dehydrated by water volume restriction. Dehydration was confirmed by body weight loss of -5.4% and -4.6% in young and old groups, respectively, and a 1.7-fold increase of kidney renin gene expression in the young rabbits. LC-MS/MS identified 2,286 proteins in the rabbit vocal folds of young and old rabbits combined. Of these, 177, 169, and 81 proteins were significantlyaffected by age, hydration status, or the interaction of both factors, respectively. Comparative proteomics revealed a higher number of proteins differentially regulated in the vocal folds of older dehydrated rabbits compared to older euhydrated, and when compared to younger rabbits, regardless of hydration status. These proteins are predominantly related to structural components of the extracellular matrix and muscle layer, suggesting a disturbance in the viscoelastic properties of aging vocal fold tissue, especially when subjected to systemic dehydration.

Project description:Vocal cord healing is a dynamic process, and many genes and proteins are involved, which play varying roles at different regeneration stages after injury. Previous studies have shown that inflammatory responses occur at the early stage of vocal cord injury, where the fibroblasts proliferate exuberantly with intensive secretion and deposition of ECM. These activities reach the peak at 3-7 days and their intensity begins to decline 15 days later. A study based on the dermal system has shown that ECM remodeling during the repair of injury can last for several months. However, few studies have been conducted as to the dynamic changes of gene expressions and signaling pathway during the healing process of vocal cord injury. Plotting these changes will facilitate the understanding about the physiological changes during healing and the identification of key time points and target genes in fibrosis formation.
