Project description:Mouse KrasG12D/P53-/-(KP) lung tumor cells were transfected with pLKO.1-Tet on-shNpm1-3(N3), pLKO.1-Tet on-shNpm1-4(N4) or pLKO.1-Tet on-shNpm1-5 (N5) and then selected by using 2ug/ml puromycin, to establish the 3 stable cell lines. These 3 stable cell lines (N3, N4 or N5) were treated with 100ng/ml Doxycline and then the total RNA of the treated and untreated cells were harvested for RNA seq.

Project description:The nucleolar scaffold protein NPM1 acts as a multifunctional regulator of cellular homeostasis, genome integrity, and stress response. We and others show that NPM1 is also involved in regulation of the autophagic pathway. As NPM1 binds to members of the GABARAP protein family, important for autophagy, we anticipate that this interaction is important for the role of NPM1 in autophagy. In order to identify NPM1-dependent GABARAPL1 interactors we performed HA-GABARAPL1 immunoprecipitation with siRNA-mediated NPM1 depletion.

Project description:Mll2 (ALR) is a histone 3 lysine 4 trimethyltransferase to function as gene activation.In our study, we found that Mll2 is vital for proper control of proliferation and lineage differentiation of mouse ESCs, particularly towards the cardiac-specific lineages. We used microarrays to detail the global programme of gene expression to compare the difference after Mll2 knockdown in E14 cell lines. E14 cells were infected with lentiviruses expressing short hairpin RNA (shRNA) for Mll2 for 24 hours, and then puromycin was added at 2mg/ml to select for 5 days. Then collet samples of control and Mll2 knockdown E14 cells for RNA extraction and hybridization on Affymetrix microarrays.
