The Holm oak (Quercus ilex) proteome as defined by shotgun MS (nLC-MS/MS, Orbitrap) approach

Project description:Holm oak (Quercus ilex) is the most important and representative specie of the Mediterranean forest and of the Spanish agrosilvo-pastoral ecosystem “Dehesa”. Despite its environmental and economic interest, Holm oak is still an orphan species whose biology is very little known, especially at the molecular level. In this research, we have performed a shotgun proteomic approach (nLC-MSMS, Orbitrap) to analyze the Holm oak proteome, using, as starting material, a pool generated by mixing equal amounts of homogenized tissue, including embryo, cotyledons (from mature acorns), and leaves and roots (from 6-month old plantlets grown in a greenhouse under environmental conditions). The proteome generated will be the bases of further studies on population variability, growth, development and responses to stresses in this species.

Proteomic response to drought and Phytophthora cinnamomi in Quercus ilex.

Project description:Biotic and abiotic stresses are predicted to be the main drivers of forest tree decline and mortality in a climatic change scenario. It occurs, for example, in Quercus ilex, the main component of the Mediterranean forest, becoming a major environmental, economic and social concern. The decline syndrome is the result of factors interrelated in time and space. Breeding, through variability and elite genotype selection, is the only plausible strategy for forest management and conservation in nondomesticated, orphan species as is the case of the genus Quercus. By using a shotgun proteomics approach, the effect and the responses to combined drought stress and pathogen (Phytophthora cinnamomi) have been analyzed in two Holm oak populations with contrasting responses to each individual stress. Proteomics data are correlated with that of plant survival, damage symptoms, leaf chemical composition and physiological phenotypes (water content and photosynthetic activity). Changes in the protein profile depended on the population, stress, and time, with some consistent upaccumulated proteins related to synthesis, primary and secondary metabolism, and cell wall are discussed.

Proteomic and Metabolomic study of the Quercus ilex response to Phytophthora cinnamomi, the main causal agent of the decline syndrome, and the identification of putative markers of resistance

Project description:Holm oak (Quercus ilex) is considered one of the major structural elements of the Mediterranean forests and the agrosilvopastoral Spanish “dehesa”, representing an outstanding example of ecological and socio-economic sustainability of forest ecosystems. The exotic pathogen Phytophthora cinnamomi is one of the most aggressive of woody species, and together drought is considered one of the main drivers of holm oak decline. The effect and responses of P. cinnamomi inoculation has been studied on the offspring of mother trees growing in declined and non-declined areas of two Andalusian populations (Cordoba and Huelva). Damage symptoms, mortality, and chlorophyll fluorescence have been evaluated in seedlings inoculated under humid and drought conditions. The effect and responses depended on the population, being more accused in Huelva than in Cordoba population. An integrative proteomic and metabolomic analysis revealed the involvement of different metabolic pathways in response to the pathogen in both populations, such as amino acid metabolism pathways in Huelva, and terpenoids and flavonoids biosynthesis in Cordoba. However, a differential response was not observed between seedlings inoculated under humid and drought conditions. A protective mechanism of the photosynthetic apparatus is launched in response to defective photosynthetic activity in inoculated plants, which seems to be more efficient in the Cordoba population. In addition, enzymes and metabolites of the phenylpropanoid and flavonoid biosynthesis pathways may confer higher resistance to Cordoba population. Some of these enzymes are proposed as markers of resilience, among which glyoxalase I, glutathione reductase, thioredoxin reductase, and cinnamyl alcohol dehydrogenase are candidates.

Project description:Drought is considered as a threat to the sustainability of Mediterranean forests and, of the agro-silvo-pastoral “dehesa” system. Holm oak (Quercus ilex L.) is one of the most representative species of these ecosystems and this abiotic stress causes a high tree mortality rate, being more accused at the seedling stage. In this study, an integrative analysis of transcriptome and proteome in leaves of Q. ilex seedlings under drought conditions was carried out to identify gene products involved in the metabolic pathways related to drought tolerant mechanisms.

Rhytidome- and cork- type barks of holm oak, cork oak and their hybrids highlight processes leading to cork formation

Project description:The periderm is basic for land plants due to its protective role during radial growth, which is achieved by the polymers deposited in the cell walls. Despite the research on the topic has unravelled the role of several enzymes and transcription factors, many questions remain open, especially those regarding cell development. Here we use the outer bark of cork oak (cork), holm oak (rhytidome), and their natural hybrids’ to further understand the mechanisms underlying periderm development. Cork is an outstanding model as it consists of a thick and very homogeneous periderm produced by a permanent mother-cell layer (phellogen). Conversely, holm oak contains a more heterogeneous bark including several thin periderms mixed with phloem, also known as a rhytidome. The inclusion of hybrid samples showing rhytidome-type and cork-type barks is valuable to approach cork development, allowing an accurate identification of candidate genes and processes. The present study underscores that biotic stress and cell death signalling are enhanced in rhytidome-type barks while lipid metabolism and cell cycle are enriched in cork-type barks. Based on the DEGs most expressed related to development, we highlight that cell division, cell expansion, and cell differentiation could account for the differences found between cork and rhytidome-type barks.

We investigated the effects of short-term N addition on soil microbial C-cycling functional gene composition, SOC-degrading enzyme activities, and CO2 emission in a 5-year field experiment established in an artificial Pinus tabulaeformis forest on the Loess Plateau, China.
