Project description:We found a heat-resistant jujube cultivar in our previous study, but the molecular mecanism of heat-resistantance remained investigated. In the current study, we made this seedlings of jujube cultivar to be under heat stress (45°C) for 0, 1, 3, 5 and 7 days respectively. After checking the phenotypic and physiological features, the leaf samples (HR0, HR1, HR3, HR5 and HR7) were collected accordingly. RNA-seq transcriptome comparisons were performed, showing that 2266, 4907, 6120 and 2894 differentially expressed genes (DEGs) were identified among HR1 vs. HR0, HR3 vs. HR0, HR5 vs. HR0, and HR7 vs. HR0 respectively. Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses of the DEGs from these comparisons. it revealed that a series of biological processes involved in photosynthesis, protein processing in endoplasmic reticulum and metabolism, suggesting that lowering or upregulating these processes may contribute to improved heat resistance in this jujube cultivar.

Project description:Jujube (Ziziphus jujuba Mill.) is an economically and agriculturally significant fruit crop and is widely cultivated throughout the world. Heat stress has recently become one of the major abiotic stresses limiting plant growth and productivity. However, there are few studies on the transcriptome profiling of jujube subjected to heat stress. In this study, we analyzed the physiological and transcriptomic changes of heat-resistant jujube cultivar ‘HR’ and heat-sensitive cultivar ‘HS’ caused by high temperature stress. We statistically determined 984, 1468, 1727 and 2098 differentially expressed genes (DEGs) between ‘HR’ and ‘HS’ after 0, 1, 3, 5 d of heat stress, respectively. Gene Ontology (GO) enrichment analysis indicated that Aa great deal of heat-responsive genes were identified in these DEGs by Gene Ontology (GO) enrichment analysis. It suggests the distinct molecular mechanism of jujube response to heat stress. Furthermore, we validated the expression profiles of 12 candidates using qRT-PCR to further confirm the accuracy of the RNA-seq data. These results will advance our knowledge of the genes involved in the complex regulatory networks of heat stress and provide genetic resources for further improving the heat tolerance in jujube.

Project description:High temperature is increasingly becoming one of the prominent environmental factors affecting the growth and development of maize (Zea mays L.). Therefore, it is critical to identify key genes and pathways related to heat stress (HS) tolerance in maize. Here, we identified a heat-resistant (Z58D) and heat-sensitive (AF171) maize inbred lines at seedling stage. Transcriptomic analysis identified 3,006 differentially expressed genes (DEGs) in AF171 and 4,273 DEGs in Z58D under HS treatments, respectively. Subsequently, GO enrichment analysis showed that shared upregulated genes in AF171 and Z58D involved in response to HS, protein folding, abiotic and temperature stimulus pathway. Moreover, the comparison between the two inbred lines under HS showed that response to heat and response to temperature stimulus significantly overrepresented for the 1,234 upregulated genes. Furthermore, commonly upregulated genes in Z58D and AF171 had higher expression level in Z58D than AF171. In addition, maize inbred CIMBL55 had been verified to be more tolerant than B73 and commonly upregulated genes had higher expression level in CIMBL55 than B73 under HS. The consistent results indicated that heat-resistant inbred lines may coordinate the remarkable expression of genes in order to recover from HS. Additionally, 35 DEGs were conserved among 5 inbred lines by a comparative transcriptomic analysis. Most of them were more pronounced in Z58D than AF171 at expression level. Those candidate genes may confer thermotolerance in maize.

Project description:In order to find out the genes involved in the heat shock response of cucumber, the seedlings of '9930', a North China type cucumber variety, were treated with high temperature at 42℃, and the leaves of the seedlings were taken after 0, 3 and 6 hours of treatment for transcriptome sequencing to analyze the differential expression genes of cucumber in response to high temperature stress. The results showed that compared with the control (heat treatment at 0h), there were 6 082 genes differentially expressed after heat treatment for 3 h and 6 h, of which 3245 were up-regulated and 2 388 were down-regulated. The results of enrichment analysis of GO and KEGG showed that these differentially expressed genes were mainly involved in metabolic pathway and protein synthesis pathway. Among the differentially expressed genes, there are many Hsp family genes, including Hsp20, HSP70 and Hsp90, which are up-regulated by heat stress, indicating that these genes may play a positive regulatory role in heat shock response of cucumber. In addition, many transcription factors, including AP2, MYB, WRKY, bHLH and HSF, were also induced by heat.

Project description:To better understand the mechanisms that regulate the heat stress response in rice, we conducted a comparative analysis of transcriptome profile in panicles from two rice lines, heat-tolerant line 252 (HTL252) and heat-susceptible line 082 (HSL082) using rice Affymetrix GeneChip. In HTL252 panicles, 1538 differentially expressed genes (DEGs) genes with at least four-fold expression changes compared with the control under heat treatment were considered heat-responsive (HR). Of these DEGs, 522 genes were up-regulated while 1016 genes were repressed. Among DEGs in HSL082, 496 genes were induced and 1707 genes were repressed. Out of the 370 common DEGs found between HTL252 and HSL082, 129 genes were induced and 241 genes were repressed.

Project description:Bacterial endospores (spores) are among the most resistant living forms on earth. Spores of Bacillus subtilis A163 show extremely high resistance to wet heat compared to spores of laboratory strains. In this study, we found that spores of B. subtilis A163 were indeed very wet heat resistant and released dipicolinic acid (DPA) very slowly during heat treatment. We also determined the proteome of vegetative cells and spores of B. subtilis A163 and the differences in these proteomes from those of the laboratory strain PY79, spores of which are much less heat resistant. This proteomic characterization identified 2011 proteins in spores and 1901 proteins in vegetative cells of B. subtilis A163.

Heat stress has recently become a primary abiotic stressor limiting the productivity and growth of jujube, as well as other crops. There are few studies, however, that have performed transcriptome profiling of jujube when it is exposed to heat stress. In this study, we observed the physiochemical changes and analyzed gene expression profiles in resistant jujube cultivar 'HR' and sensitive cultivar 'HS' subjected to heat stress for 0, 1, 3, and 5d. Twenty-four cDNA libraries from 'HR' and 'HS' leaves were built with a transcriptome assay. A total of 6887 and 5077 differentially expressed genes were identified in 'HR' and 'HS' after 1d, 3d, and 5d of heat stress compared with the control treatment, GO and KEGG enrichment analysis revealed that some of the genes were highly enriched in oxidation-reduction process, response to stress, response to water deprivation, response to heat, carbon metabolism, protein processing in endoplasmic reticulum, and plant hormone signal transduction and may play vital roles in the heat stress response in jujube plants. Differentially expressed genes were identified in the two cultivars, including heat shock proteins, transcriptional factors, and ubiquitin-protein ligase genes. And the expression pattern of nine genes was also validated by qRT-PCR. These results will provide useful information for elucidating the molecular mechanism underlying heat stress in different jujube cultivars.

Project description:Background: Plant disease is a major challenge to agriculture worldwide, and it is often exacerbated by abiotic environmental factors. During some plant-pathogen interactions, heat stress increases host susceptibility, a tendency which could spell disaster in light of the global warming trends associated with climate change. Despite the importance of this phenomenon, little is known about the molecular mechanisms that cause it. To better understand host plant responses during simultaneous heat and pathogen stress, we conducted a transcriptomics experiment for rice plants infected with Xanthomonas oryzae (Xo), an economically important bacterial pathogen of rice, during high temperature stress. Results: Using RNA-Seq technology, 8,499 differentially expressed genes were identified as temperature responsive in one rice cultivar, IRBB61, experiencing susceptible and resistant interactions with Xo across three time points. Many genes with gene ontology terms associated with stress response were identified. Notably, genes in the plant hormone abscisic acid (ABA) biosynthesis and response pathways were identified as upregulated by high temperature in both mock-treated plants and plants in the susceptible interaction and suppressed by high temperature in plants in the resistant interaction. A DNA sequence motif similar to known ABA-responsive cis-regulatory elements was identified in the promoter region upstream of genes upregulated in susceptible but downregulated in resistant interactions. Conclusions: The results of our study suggest that the plant hormone ABA is an important node for cross-talk between plant transcriptional response pathways to high temperature stress and pathogen attack. Genes in this pathway represent an important focus for future study to determine how plants evolved to deal with simultaneous abiotic and biotic stresses.
