Transcriptome analysis of MCF-7 breast cancer cells in which VASP gene was knocked down

Project description:Previous studies have shown that the main function of VASP is to regulate the cytoskeleton and play an important role in promoting tumor cell metastasis. In this study, we first reveal that VASP is located in the nucleus of breast cancer cells and elucidate a Wnt/β-catenin/VASP positive feedback loop. We identify that VASP is a target gene of Wnt/β-catenin signaling pathway, and activation of Wnt/β-catenin signaling pathway can significantly upregulate VASP protein expression, while upregulated VASP protein can in turn promote translocation of β-catenin and DVL3 proteins into the nucleus. In the nucleus, VASP, DVL3, β-catenin and TCF4 can form VASP/DVL3/β-catenin/TCF4 protein complex, activating Wnt/β-catenin signaling pathway, and promoting the expression of target genes VASP, c-myc and cyclin D1. Thus, our study reveals that there is a Wnt/β-catenin/VASP malignant positive feedback loop in breast cancer, which promotes the proliferation and migration of breast cancer cells, and breaking this positive feedback loop may provide new strategy for breast cancer treatment.

Project description:CD24 is the one of cell surface protein that anchored via glycosyl phosphatidylinositol that links to cell surface and modulates growth and differentiation signal many of cell types. A small population of cells, namely ‘Cancer Stem Cells (CSCs)’, charges highly aggressive and metastatic character of cancer cells and shows conformational change of ‘epithelial to mesenchymal transition (EMT). To understand the role of CD24 in CSC and EMT, CD24 was knocked down using siRNA in the MCF7 cell line (MCF-7 hCD24 KD) and analyzed transcriptome profiles by mRNA-sequencing. Isobaric tags for relative and absolute quantification based on quantitative proteomic analysis on inhibitor of apoptosis protein-like protein-2 knocked-down MCF-7 breast cancer cells

Project description:Although the inhibitor of apoptosis protein-like protein-2 (ILP-2) is regarded as a novel growth enhancer for breast cancer, its mechanism on breast cancer cell growth remains elusive and requires further study. In this study, isobaric tags were applied for relative and absolute quantification (iTRAQ) analysis on two groups of MCF-7 (Michigan Cancer Foundation-7) breast cancer cells, namely, siRNA-5 group (knocked down, KD group) and negative control (NC) group, to analyse the protein expression profiles correlated to ILP-2 during breast cancer cell growth. Western blot was applied to verify the iTRAQ data. The results indicated that a total of 4065 proteins were identified in breast cancer cells, and 241 of these proteins are differentially expressed (fold change ≥ 1.20 or ≤ 0.83 and P<0.05). A total of 156 up- and 85 down-regulated proteins are found in siRNA-5 group versus NC group. These differentially expressed proteins are principally correlated to the ECM (extracellular matrix) -receptor interaction, and the proteins from the top 10 biological processes are correlated to signal transduction, regulation of cell proliferation and immune system processes. The expression of AGA (N(4)-(beta-N-acetylglucosaminyl)-L-asparaginase), MT1E (metallothionein-1E) and TDO2 (tryptophan 2,3-dioxygenase) increases when the protein expression of ILP-2 is knocked down. These results suggested that ILP-2 has an important role in breast cancer cell growth via protein interaction and provided comprehensive insights into biochemical pathways and regulation networks correlated to ILP-2.
