Project description:Aberration in miRNA expression or DNA methylation is a causal factor for numerous pathological conditions including polycystic ovarian syndrome PCOS, a common endocrine disorders and leading cause of infertility. The epigenetic interactions between miRNA and DNA methylation remain unexplored in PCOS. Few studies have reported that the regulation of miRNAs in polycystic ovary syndrome(PCOS). Our study helps to understand the molecular pathogenesis of PCOS in human ovarian granulosa cells from the perspective of post-transcriptional level.

Expression profile of plasma exosomal microRNAs in women with polycystic ovary syndrome and control women

Project description:Polycystic ovary syndrome (PCOS) is a highly prevalent endocrine-metabolic disorder. Epigenetic etiology is suggested to play an important role in PCOS pathogenesis, since it is hard to elucidate completely by genetic variations. Exosomal microRNAs (miRNAs) have recently been identified as a novel mechanism of intercellular communication, and a more stable regulator of gene expression compared with free miRNAs. The aim of this study is to detect the different expression profile of plasma exosomal miRNAs between PCOS patients and healthy controls.

Identification of exosomal miRNAs specifically derived from intrafollicular cells involved in polycystic ovary syndrome

Project description:Exosomes have recently been shown to play a key role in cell-to-cell communication through delivery of various functional content, including microRNAs (miRNAs). We investigated the potential roles of exosomal miRNA derived intrafollicular cells in polycystic ovary syndrome (PCOS). Using microarray profiling, a total of 492 miRNAs and 220 miRNAs were found in follicular fluid-derived exosomes and serum-derived exosomes, respectively, in PCOS and non-PCOS females. By excluding miRNAs existing in serum-derived exosomes, we found 179 miRNAs which were specifically expressed in follicular fluid-derived exosomes both in PCOS and non-PCOS females. Using microarray profiling, a total of 492 miRNAs and 220 miRNAs were found in follicular fluid-derived exosomes and serum-derived exosomes, respectively, in PCOS and non-PCOS females.
