Project description:Our goal is to discriminate specific genes in live M.leprae-infected peritoneal macrophages in comparison to heat-killed M.leprae infected peritoneal macrophages using microarray. Two-condition experiment, Heat-killed M.leprae infected macrophage vs. Live M.leprae infected macrophage. Biological replicates: 16 mice (control), 8 heat-killed M. leprae mice (sample 1), and 8 live M. leprae infected mice (sample 2). Independently grown and harvested from isolator. One replicate per array.

Heat-killed probiotic Levilactobacillus brevis MRKAK9 downregulates insulin-signaling pathway and improves proteostasis and autophagy to promote longevity and immunity in Caenorhabditis elegans

Project description:Probiotics play important role in maintaining the health and extend longevity in their host. Previous studies reported several live probiotic bacteria in enhancing longevity and improving diverse feature of the host’s health. In this study, we reported a new potential heat- killed probiotic bacterium Levilactobacillus brevis strain MRKAK9 improved longevity and different features of healthy aging, including age-associated physical activity, improved resistance to biotic and abiotic stress in C. elegans. The mechanistic investigations showed that heat-killed strain MRKAK9 promoted longevity and healthy aging by downregulating insulin-signaling pathway resulting in improved proteostasis, autophagy and preserving lysosomal functionality in C. elegans. Heat-killed strain MRKAK9-mediated downregulation of insulin signaling pathway was regulated by the miRNA mir-243, suggesting that mir-243 partially involved in enhancing longevity of C. elegans. Additionally, the structural component exopolysaccharide of the strain heat-killed MRKAK9 also downregulated insulin- signaling mechanisms, increased the expression of genes involved in proteostatsis, autophagy as well as improved expression of mir-243. This study indicates that heat-killed strain MRKAK9 improves longevity and healthy aging, suggesting its candidature as a novel postbiotic.

RNA-seq analysis of Fusarium graminearum Z-3639 in heat stress

Project description:Fusarium graminearum (teleomorph Gibberella zeae) is a prominent pathogen that infects major cereal crops, such as wheat, barley, and maize. To dissect cellular responses toward heat stress in the plant pathogenic fungus F. graminearum, we compared transcriptomes of the fungal cultures incubated in normal temperature condition (25 ÂºC) and in high temperature condition (37 ÂºC) for 15 min. 6 samples examined: 24 h-old mycelia from complete medium (CM) of Fusarium graminearum wild-type Z-3639 were incubated in normal temperature condition (25 ÂºC) and in high temperature condition (37 ÂºC) for 15 min.

Drosophila melanogaster S2 cells nucleosome occupancy data after simulation by heat killed Salmonella typhimurium

Project description:In eukaryotes, nucleosomes participate in all DNA-templated events by regulating access to the underlying DNA sequence. However, the dynamics of nucleosomes during a genome response has not been well characterized . We stimulated DrosophilaM-BM- S2 cells with heat-killed Gram-negative bacteria Salmonella typhimurium, and mapped genome-wide nucleosome occupancy at high temporal resolution by MNase-seq using Illumina HiSeq 2500. We show widespread nucleosome occupancy change in S2 cells during the immune response, with the biggest nucleosomal loss occurring at 4hr post stimulation.
