Project description:Altogether, this is the first study presenting the global changes of the transcriptome in the adrenal microvascular cells. The results of RNA sequencing and subsequent bioinformatics analysis revealed that adrenal vascular endothelial cells are active players not only involved in maintenance of the adrenal gland microenvironment, but also they play a key role in pathogen recognition and initiation of inflammation. However, our results also strongly suggest that systemic administration of LPS, which recapitulates to some degree situation in people with meningococcal sepsis, may develop certain damage, including vascular inflammation, hypoxia, blood coagulation and leakage. That may contribute to the adrenal dysfunction.

Project description:Although of the utmost importance for our survival, the activation of the adrenal gland is often impaired during sepsis. Little is known about the mechanisms involved in this process. In the present study, we have used a robust RNA Sequencing (RNA-Seq) technology in a search for novel transcriptomic signatures in adrenal glands triggered by bacterial lipopolysaccharide-induced systemic inflammation. Our results demonstrate that LPS injection to mice induce a robust change in the adrenal transcriptome, significantly altering the expression of 8458 genes as compared to saline injected animals. The principal component analysis shows a clear separation between LPS and saline treatments, as well as show low variance between biological samples from each group. The functional analysis of 4312 upregulated and 4146 downregulated genes by LPS, demonstrate a strong induction of genes involved in inflammatory and steroidogenic responses, and inhibition of genes involved in DNA repair and stem/progenitor cell pathways. Gene enrichment set analysis (GSEA) further validated the functional data, and indicated a strong grouping of genes involved in LPS-mediated induction of interferon and innate immune responses, along with previously unexplored activation of the hypoxia and inhibition of Wnt/Shh pathways in the adrenal gland. These results were further validated by quantitative PCR analysis. Collectively, this is the first study demonstrating a global alteration of adrenal gene signatures during systemic inflammation, indicating the potential pathways that are involved in the adrenal gland dysregulation during sepsis.

Previously, LPS challenge in the male piglets of these sows resulted in differences in fever and cortisol responses among piglets from different maternal treatments. Project description:Genipin is a natural blue colorant in food industry. Inflammation is correlated with human disorders, and nuclear factor-κB (NF-κB) is the critical molecule involved in inflammation. In this study, the anti-inflammatory effect of genipin on the lipopolysaccharide (LPS)-induced acute systemic inflammation in mice was evaluated by NF-κB bioluminescence-guided transcriptomic analysis. Transgenic mice carrying the NF-κB-driven luciferase genes were administered intraperitoneally with LPS and various amounts of genipin. Bioluminescent imaging showed that genipin significantly suppressed LPS-induced NF-κB-dependent luminescence in vivo. The suppression of LPS-induced acute inflammation by genipin was further evidenced by the reductions of cytokine levels in sera and organs. Microarray analysis of these organs showed that the transcripts of 79 genes were differentially expressed in both LPS and LPS/genipin groups, and one third of these genes belonged to chemokine ligand, chemokine receptor, and interferon (IFN)-induced protein genes. Moreover, network analysis showed that NF-κB played a critical role in the regulation of genipin-affected gene expression. In conclusion, we newly identified that genipin exhibited anti-inflammatory effects in a model of LPSinduced acute systemic inflammation via downregulation of chemokine ligand, chemokine receptor, and IFN-induced protein productions. A total of 25 transgenic mice (female, 6 to 8 weeks old) were randomly divided into five groups of five mice: (1) mock, no treatment; (2) LPS (4 mg/kg), (3) LPS plus genipin (1 mg/kg), (4) LPS plus genipin (10 mg/kg), and (5) LPS plus genipin (100 mg/kg). Mice were challenged intraperitoneally with LPS and then with genipin 10 min later. Four hours later, mice were imaged for the luciferase activity, and subsequently sacrificed for ex vivo imaging, RNA extraction, and immunohistochemical staining.

Project description:Endothelial dysfunction is a hallmark of LPS-induced acute kidney injury (AKI). Endothelial cells (EC) acquired a fibroblast-like phenotype and contributed to myofibroblasts generation through Endothelial to Mesenchymal Transition (EndMT) process. Noteworthy, ARPCs enhance tubular regenerative mechanism during AKI, but little is known about their effects on EC. Here we investigated whether ARPCs could prevent sepsis-induced EndMT and the related mechanism. When activated by LPS, Human endothelial cells (EC) proliferated and decreased specific EC markers such as CD31 and VE-cadherin and up-regulated myofibroblast markers such as Collagen I and Vimentin. The co-culture with ARPCs normalized EC proliferation rate and abrogated the LPS-induced EndMT by restoring the high expression of EC markers and the low expression of myofibroblast markers. Gene set enrichment analysis showed that most of genes modulated in LPS-stimulated ARPCs belongs to cell activation and defense response pathways. In particular, among most up-regulated genes we found BPIFA2, SAA2, SAA4 and CXCL6. BPIFA2 is recently described as an early biomarker of AKI but little is known about its function in the kidney. The other genes are frequently involved in the response to bacterial infection and kidney injury. Finally, in a swine model of LPS-induced AKI, we observed an increase of CD133+ARPCs that expressed BPIFA2 respect to healthy pigs. Taken together, these results suggest an underestimate role of ARPCS in preventing endothelial dysfuncton by the production of several proteins. The identification of these molecules may offer novel strategies to protect endothelial compartment and promote kidney repair.
