Influence of Microbial and Host Cell Sublethal Heat Stress on S. Typhimurium Gene Expression

Project description:Environmental stress contributes to the outcome of infection by impacting both microbial virulence and host susceptibility to infection. Thermal processing, commonly used for decontamination of poultry in the food industry, may elicit sublethal stress on resistant serovars of Salmonella. We employed traditional heat shock temperatures (42 and 48ºC), similar to avian body temperature and poultry processing conditions, to study gene expression of Salmonella enterica serovar Typhimurium. Microarray analysis indicated that thermal shock at 42°C and 48°C induced expression of SPI-2 and SPI-5 genes, whose products are required for adhesion and survival. However, SPI-1 genes, responsible for invasion of Salmonella into host cells, were down-regulated following exposure to 42°C and 48°C. Bacterial adhesion assays showed greater adhesion of heat-stressed S. Typhimurium to Caco-2 cells compared to non-stressed bacteria. In addition, subjecting Caco-2 cells to mild heat shock (39°C), which is similar to human fever, enhanced host cell susceptibility to bacterial adhesion. Data indicate that thermal stress enhances bacterial colonization and host cell susceptibility to adhesion during S. Typhimurium infection.

RNAseq of a chicken lung epithelial cell line (cLEC-213) upon treatment with butyrate and/or infection with low pathogenic avian influenza

Project description:Short-chain fatty acids (SCFAs) are the end-products of bacterial fermentation of dietary fibre, and can play a crucial role in regulating immunity and metabolism in the gut and peripheral tissues. Here we show that butyrrate, an SCFA, displayed antiviral effects in chicken respiratory epithelial cells, likely via the regulation of interferon-stimulated genes, particularly OASL. The observed antiviral signaling mechanism would involve HDAC inhibition and Sp1-dependent regulation of the OASL promoter, shedding light on new mechanisms through which the GM regulates poultry mucosal immunity along the gut-lung axis in the chicken.

Effects of Escherichia coli global gene expression due to microfluidic processing

Project description:Investigation in bacterial transcriptomics is widely used to investigate gene regulation, bacterial susceptibility to antibiotics, host-pathogen interactions, and pathogenesis. Transcriptomics is crucially dependent on suitable methods to isolate and detect bacterial RNA. Microfluidic approaches offer ways of creating integrated point-of-care systems, analysing a sample from preparation, RNA isolation through to detection. Critical for on-chip diagnostics to deliver on their promise is that mRNA expression is not altered through the use microfluidic sample processing. Here, we investigate the impact on the use of a microfluidic sample processing system based on hydrodynamic separation upon RNA expression of bacteria isolated from blood to prove its suitability for further microfluidic test development.

Project description:Bacterial infections remain an important cause of morbidity in poultry production. The molecular characteristics and dynamic changes in immune cell populations after bacterial infection have yet to be fully understood. Beijing-You chicken and Cobb broiler, two broiler breeds with different disease resistance, were infected with Salmonella typhimurium, and inflammation models were constructed. We characterized chicken spleen CD45+ immune cells by single-cell RNA sequencing.
