Project description:We constructed E. coli transcriptome under deoxynivalenol (DON) and nivalenol (NIV) condition from Fusarium spp. To identify differentially expressed genes in mycotoxin condition, we compared mycotoxin (DON and NIV) transcriptome to acetonitrile (ACN) transcriptome as the solvent of myxotoxin. As a result, 435-929 transcripts were identified from all conditions, respectively.

Project description:Upon axenic cultivation in presence of the mycotoxin inducing nitrogen source L- ornithine the HEP1 deletion mutant showed an altered secondary metabolite profile including reduced levels of deoxynivalenol (DON). This finding was contrasted with a 1.5 fold increased infection rate on the susceptible wheat cv. Remus which was accompanied by increased production of DON. Transcriptome analysis of the HEP1 deletion versus the PH-1 wildtype strain during pathogenic growth state as well as during saprophytic growth on dead (non-responding) wheat heads and axenic samples allows to distinguish gene response of the pathogen reacting on signals from the active, defending plant from those regulated by plant substrate effects or in vitro mimicked mycotoxin inducing conditions, providing insights into gene regulation underlying the observed hypervirulence.

Project description:Morphology together with the capability to respond to surrounding stimuli are the key elements governing the spatial interaction of a living cell with the environment. In this respect, biomechanical stimulation can trigger significant physiological cascades that can potentially modulate or interact with toxicity. Deoxynivalenol (DON, vomitoxin) is one of the most prevalent mycotoxins produced by Fusarium spp. and it was used to explore the delicate interaction between biomechanical stimulation and cytotoxic processes in A431 cells. In fact, in addition of being a well-known food contaminant, DON is becoming a relevant toxin also for other organ systems. The combination between biomechanical stimulation and the mycotoxin revealed how DON can impair crucial functions like cellular morphology and lysosome trafficking at concentrations far below those known to be cytotoxic in routine toxicity studies. Moreover, sub-toxic concentrations of DON (0.1-1 μM) impaired the capability of A431 cells to respond to a biomechanical stimulation that normally sustains trophic effects in this cell line. Ultimately, the effects of DON (0.1-10 μM) were partially modulated by the application of uniaxial stretching (0.5Hz, 24h, 15% deformation) suggesting a deep cross-talk between the cytotoxic potential of the mycotoxin and the biomechanical stimulation.

The diets consisted of a control diet and of three diets with moderate concentrations of 1.0, 2.5 and 5.0 mg DON/kg feed, which was attained by exchanging uncontaminated wheat with naturally DON-contaminated wheat up to the intended concentration. The chicken were held at standard conditions for 23 days and received their diet ad libitum. After slaughter the gene expression was determined in the liver of three samples per group.

Among animal species, pigs are the best model for humans and are very sensitive to DON. In wheat, DON can be conjugated to glucose to form DON-3-β-D-glucoside (D3G). Some bacteria isolated from digestive tracts or soil, are also able to de-epoxydize or epimerize DON to metabolites such as deepoxy-deoxynivalenol (DOM-1) or 3-epi-deoxynivalenol (epi-DON). The toxicity of these DON metabolites is poorly documented. By the way of ingestion, the intestine is the first organ exposed to these molecules and so constitute a relevant model. The aim of this study was to compare the intestinal toxicity of three DON metabolites (D3G, DOM-1 and epi-DON) with the one of DON. Intestinal explants from 6 pigs were treated with 10mM DON, D3G, DOM-1 or epi-DON for 4 hours and transcriptomic analysis was performed using an “Agilent Porcinet 60K”.

Project description:The effect of the mycotoxins deoxynivalenol (DON) and zearalenone (ZEN) on the gut microbiome of 15 weaners were analysed. Animals were fed for 28 days with low and high dosages of the toxins. Animals of the control group did not receive any mycotoxin-spiked feed. The gut metaproteome composition shifted due to the high dosages of the toxins compared to the control and low mycotoxin groups and it was also more similar between the animals that received high dosages of the mycotoxins.

Project description:Fusarium graminearum (F. graminearum) and its derivative mycotoxin deoxynivalenol (DON) are highly concerned with food security, safety and sustainability worldwide. The molecular mechanism regulates DON biosynthesis in F. graminearum remains enigmatic, despite several transcription factors (TFs) have been revealed containing regulatory functions. Here, we first characterized a zinc finger-contained TF, FgSfp1. Interestingly, contrast to the previous knowledge, all TRI-cluster genes were abnormally upregulated in ∆FgSfp1 while Tri proteins abundance rationally decreased, resulting in a 95.4% reduction of DON yield simultaneously. Further evidence determined FgSfp1 acted as a nutrient-sensing factor coordinates genetic expression efficiency through interference of ribosomal biogenesis process. Specifically, FgSfp1-depletion leads to ribosome biogenesis assembly factor (RiBi) expression attenuation along with DON precursor acetyl-CoA synthase reduction since FgSfp1 actively interacts with RNA 2’-O-methylation enzyme FgNop1 revealed by Bi-FC and subsequently influences mRNA translation capacity. In conclusion, we elucidated that the FgSfp1 orchestrates DON biosynthesis via participating RNA posttranscriptional modification for ribosomal RNA maturation, offering new insights into the DON biosynthesis regulation. Ultimately, this novel TF might be a key regulator for DON contamination control in the whole food chain.
