Project description:We combined an experimental microbiome of 11 bacterial strains isolated from the gut of native Caenorhabditis elegans. C. elegans were maintained on the experimental microbiome, Escherichia coli OP50 (control food source), or OP50 supplemented with cell-free media (CFM) from the experimental microbiome. For each of the three feeding conditions, RNA-seq was performed for wildtype (N2) worms or transgenic worms expressing amyloid beta 1-42 in their body wall muscle (GMC101).

Project description:Transcriptional profiling of C. elegans young adult worms cultured on non-pathogenic Bacillus strain 67 versus versus age-matched worms cultured on the control lab food E. coli OP50. The goal was to identify genes regulated in response to differences in diet, which might confer immunity to later exposures to pathogenic Bacillus thuringiensis DB27. One-condition experiment. C. elegans young adults, cultured on : Bacillus strain 67 versus E. coli OP50. 4 biological replicates, including 2 dye-swaps.

Caenorhabditis elegans infection with Candida albicans

Project description:N2 young adult animals were analyzed four hours after exposure to wild-type Candida albicans DAY185, heat-killed C. albicans DAY185 and heat-killed Escherichia coli OP50, all on Brain Heart Infusion (BHI) agar. It was necessary to use heat-killed E. coli OP50 as a control for these experiments because live E. coli OP50 (the normal nematode food source) is pathogenic to nematodes on BHI agar. These data identify the C. elegans genes that are differentially regulated during nematode infection with a human fungal pathogen.

Gene expression by microarray of two genotypes of Caenorhabditis elegans carrying human alpha-synuclein

Project description:We conducted a timeseries experiment on Caenorhabditis elegans genotypes NL5901 and SCH4856, which carry a human alpha-synuclein gene in an N2 and a CB4856 genetic background respectively. We grew synchronized populations of the nematodes under normal conditions (20 degrees Celcius, feeding on Escherichia coli OP50) for 24 to 216 hours. Each 24 hours plates were sampled. The goal of the experiment was to identify a timepoint where the differential development speed of the two genotypes had little influence on gene expression profiles.

The broader goal of study was to study evolution of pathogen response by comparing this expression profile to that obtained by exposing the nematode Pristionchus pacificus to the same pathogen. Other experiments which are a part of this study include expression profiling of C. elegans and P. pacificus on other pathogens including Bacillus thuringiensis, Staphylococcus aureus, and Xenorhabdus nematophila.
